Versatile Types of Inorganic/Organic NIR-IIa/IIb Fluorophores: From Strategic Design toward Molecular Imaging and Theranostics

Summary: In vivo imaging in the second near-infrared window (NIR-II) has provided great opportunities for biomedical research and clinical applications. This review summarizes the design, properties, molecular imaging, and theranostics of inorganic/organic NIR-IIa/IIb fluorophores. The versatile applications and imaging modalities enabled by these advanced fluorescent properties are discussed, along with the challenges and perspectives for future clinical translation to boost the progress of NIR-IIa and NIR-IIb imaging technology.

By utilizing different emission properties of the products, ONOO- levels can be monitored in the green channel and ATP concentrations in the red channel. The probe was successfully used in hepatocytes to demonstrate changes in signal intensity in response to ATP synthase inhibition and exogenous ONOO- exposure.

Rational Design and Application of an Indolium-Derived Heptamethine Cyanine with Record-Long Second Near-Infrared Emission

Summary: In this work, a novel indolium-like heptamethine cyanine dye was designed and developed by installing indolium-derived polycyclic aromatic hydrocarbons on a conjugated polyene backbone. This dye showed bright fluorescent emission in the second near-infrared (NIR-II) window and had the capability of marking blood vessels in mice. Moreover, it exhibited strong absorption bands in the NIR range and high photothermal conversion efficiency, making it suitable for tumor-targeted photothermal therapy.

Conjugated Oligoelectrolytes for Long-Term Tumor Tracking with Incremental NIR-II Emission

Summary: The design and synthesis of a near-infrared (NIR)-II emissive conjugated oligoelectrolyte COE-BBT is reported. COE-BBT exhibits high solubility in aqueous media, low toxicity, and a propensity to intercalate lipid bilayers. It provides two emission channels, facilitating in vitro confocal microscopy and in vivo animal imaging. The NIR-II emission of COE-BBT is used to track tumor progression in mice.

A General Strategy for Development of Activatable NIR-II Fluorescent Probes for In Vivo High-Contrast Bioimaging

Summary: Organic dye based NIR-II fluorescent probes offer high signal-to-background ratio and deep-tissue imaging capabilities. A novel class of polymethine dyes, NIRII-RTs, with bright and stable NIR-II emission and large Stokes shifts have been designed. These dyes, particularly NIRII-RT3 and NIRII-RT4, can serve as effective platforms for activatable bioimaging probes. Target-activatable NIRII-RT probes for pH, ATP, and metal-ion detection were synthesized, demonstrating real-time monitoring of drug-induced hepatotoxicity.

Among these probes, WH-3 exhibited high sensitivity towards H2S and enabled in vivo tracking and imaging of H2S with high contrast. This molecular design strategy shows promise in developing activatable NIR-II fluorescent probes with long emission.

Xanthene-Based NIR-II Dyes for In Vivo Dynamic Imaging of Blood Circulation

Summary: This study introduces a series of xanthene-based NIR-II dyes, named VIXs, with VIX-4 showing the best performance in terms of fluorescence emission at 1210 nm and high brightness. The dye has been successfully used for dynamically imaging the blood flow of mice, allowing for direct differentiation between arteries and veins and measurement of blood flow volume through videos. The study not only provides an effective tool for high spatial and temporal resolution bioimaging, but also presents a new promising conjugated skeleton for NIR-II dyes.

Tailored Engineering of Novel Xanthonium Polymethine Dyes for Synergetic PDT and PTT Triggered by 1064 nm Laser toward Deep-Seated Tumors

Summary: In this study, five novel NIR-II dyes were successfully developed with intense absorption ranging from 890 to 1206 nm, strong singlet oxygen generation capability, and fluorescence imaging ability triggered by a single NIR-II light. Among them, BH 1024 showed the best performance in singlet oxygen generation, moderate photothermal heating, and considerable fluorescence under 1064 nm laser irradiation. Encapsulating BH 1024 into folate-functionalized polymer demonstrated a synergetic PDT/PTT effect in vitro and in vivo, eventually achieving solid tumors elimination under NIR-II fluorescence guide. This is the first attempt to explore and design small molecular dyes for NIR-II PDT or NIR-II PDT/PTT.

Harnessing α-L-fucosidase for in vivo cellular senescence imaging
