Application of W-band 19F electron nuclear double resonance (ENDOR) spectroscopy to distance measurement using a trityl spin probe and a fluorine label

Summary: In this study, the suitability of high-field ENDOR spectroscopy in the W-band for triarylmethyl and fluorine label pairs was investigated using newly synthesized model compounds. The results showed that strong magnetic fields allowed for distinguishing nuclear frequencies of F-19 and protons with sufficient resolution. The study also applied this approach to investigate the thiol exchange process between triarylmethyl-labeled and F-19-labeled human serum albumin molecules.

Summary: Ribonucleotide reductases (RNRs) play a key role in DNA replication and repair by catalyzing the reduction of ribonucleotides. In Escherichia coli, RNRs undergo a multistep radical transfer process involving proton-coupled electron transfer. Advanced EPR spectroscopy has been used to study this mechanism and revealed important insights into the behavior of the RNR enzyme complex.

Through-Space Scalar 19F-19F Couplings between Fluorinated Noncanonical Amino Acids for the Detection of Specific Contacts in Proteins

Summary: In this study, it was demonstrated that fluorine atoms in noncanonical amino acids positioned in the hydrophobic core or on the surface of a protein can be linked by scalar through-space F-19-F-19 couplings, even if the F-19 spins are separated by more than the van der Waals distance. The use of fluorinated amino acids and F-19-F-19 interactions offer a straightforward way to probe the spatial proximity of selected sites in proteins, without requiring H-1 NMR resonance assignments.

A Chiral Lanthanide Tag for Stable and Rigid Attachment to Single Cysteine Residues in Proteins for NMR, EPR and Time-Resolved Luminescence Studies

Summary: The study introduces a new stable chiral Ln(III) tag, known as C12, for site-specific attachment to proteins, enabling protein probing through various spectroscopic techniques. The C12 tag exhibits enhanced reactivity towards selenocysteine, offering a pathway for selectively tagging selenocysteine in proteins containing cysteine residues. With Tb-III or Tm-III ions, the C12 tag can generate pseudocontact shifts in protein NMR spectra, providing a rigid tether between lanthanide and protein for interpretation of the shifts and distance measurements in double electron-electron resonance experiments. Additionally, the Tb-III complex upon reaction with cysteine or thiol compounds shows a significant enhancement in luminescence quantum yield, making it a sensitive luminescence probe for time-resolved FRET assays and enzyme reaction monitoring.
