Structural basis of Integrator-mediated transcription regulation

Isaac Fianu et al.

Summary: Integrator and PP2A collaboratively form a complex that dephosphorylates paused Pol II and cleaves nascent RNA to facilitate transcription termination. Integrator binds Pol II and pausing factors to exclude elongation factors, and positions PP2A to counteract Pol II phosphorylation. Additionally, Integrator endonuclease cleaves RNA near the Pol II active site, enabling DNA release and termination of transcription.

The PP2A-Integrator-CDK9 axis fine-tunes transcription and can be targeted therapeutically in cancer

Stephin J. Vervoort et al.

Summary: CDK9 regulates gene expression by controlling the pausing checkpoint during the transcription cycle, while PP2A counteracts CDK9-mediated phosphorylation. Loss of INTS6 leads to resistance to CDK9 inhibition-induced tumor cell death, and pharmacological activation of PP2A in combination with CDK9 inhibition provides therapeutic benefits in vivo.

Nitroxide spin labels and EPR spectroscopy: A powerful association for protein dynamics studies

F. Torricella et al.

Summary: SDSL-EPR technique involves attaching a paramagnetic label to specific positions of proteins for EPR spectroscopic study, providing information on local conformational dynamics, distances between spin centers, structures, and functions. The versatility of this spectroscopic approach in protein structural characterization has been demonstrated through recent studies, aiming to spread its applicability in various fields of structural biology, particularly for non-experts.

Targeting of SET/I2PP2A oncoprotein inhibits Gli1 transcription revealing a new modulator of Hedgehog signaling

Iliana Serifi et al.

Summary: The oncoprotein SET/I2PP2A has been identified as a novel regulator of the Hedgehog signaling pathway, showing involvement in cell proliferation, differentiation, and tumorigenesis. Experimental data support the role of SET in Gli1-mediated transcription, suggesting it as a new modulator of Hedgehog signaling.
