Auranofin: Past to Present, and repurposing

Summary: Auranofin (AF), a gold compound used for treating rheumatoid arthritis for over 40 years, has unknown mechanism of action. Research has shown that AF inhibits the induction of proinflammatory proteins and activates proteins involved in oxidative stress, but the exact transport mechanisms into and out of cells are not fully understood. Ongoing trials are exploring the repurposing of AF for treating cancer and infectious diseases, including COVID-19. Novel gold compounds are also being developed as anti-cancer and anti-infection agents.

Targeting Redox Metabolism in Pancreatic Cancer

Summary: Cell metabolism in cancer cells is reprogrammed to meet their high energy and biosynthetic demands, leading to alterations in redox metabolism and accumulation of reactive oxygen species (ROS). Cancer cells adapt to high ROS levels, contributing to tumorigenesis, metastasis, therapy resistance, and relapse. Combining strategies to increase ROS production and inhibit antioxidant capacity is a promising approach for pancreatic cancer therapy.
