Targeting tumor innervation: premises, promises, and challenges

Summary: High intratumoral nerve density is associated with poor survival, metastasis, and recurrence in solid tumors. Cancer cells release neurotrophic factors and exosomes to promote tumor innervation, and infiltrating nerves can mediate tumor biological processes through exosomes and neurotransmitters. This review discusses the communication between peripheral nerves and tumor cells in the tumor microenvironment, and the regulatory mechanisms of nerve-tumor interaction on oncogenic processes, angiogenesis, lymphangiogenesis, and immunity. Furthermore, the potential of repurposing drugs to target tumor microenvironment communication is explored, providing a new approach to prevent tumor malignancy.
