Clinical diagnosis of metabolic disorders using untargeted metabolomic profiling and disease-specific networks learned from profiling data

Summary: Untargeted metabolomics is a global molecular profiling technology used for screening inborn errors of metabolism. The study aimed to improve manual diagnosis of IEMs by developing novel computational methods for analyzing untargeted metabolomics data. The automated computational diagnostic method CTD was employed to connect metabolite perturbations and disease-specific metabolite co-perturbation networks. The study showed that CTD-based network-quantified measures can accurately diagnose various IEMs.

Summary: This study applies a computational scheme to model the recovery dynamics of severe COVID-19 and reveals the cellular and molecular mechanisms underlying the recovery process. The decrease in mature neutrophils during recovery is found to have a direct impact on disease outcome, and global regulatory changes in gene programs are observed, decoupled from cell compositional changes. The findings present a clinically relevant computational framework for modeling disease recovery and provide insights for future studies on the recovery dynamics in other diseases and tissues.

Additionally, circulating mtNFPs induce neutrophil activation through FPR1-dependent mechanisms, indicating FPR1 as a potential therapeutic target for RA.

Mapping the human genetic architecture of COVID-19

A global network of researchers identified 13 significant genetic loci associated with SARS-CoV-2 infection and severe COVID-19 cases. Collaboration among human genetics researchers has allowed for the discovery of novel host genetic factors linked to COVID-19.
