Physical properties of the cytoplasm modulate the rates of microtubule polymerization and depolymerization

Arthur T. Molines et al.

Summary: The concentration of the cytoplasm has a linear and inverse relationship with the rates of microtubule polymerization and depolymerization, indicating that changes in cytoplasmic viscosity play a key role in modulating these reactions.

Narrow-Gap Rheometry: A Novel Method for Measuring Cell Mechanics

Khawaja Muhammad Imran Bashir et al.

Summary: The viscoelastic properties of a cell cytoskeleton provide information about the cell state. Chemical agents that interact with the cytoskeleton can change cell mechanics and inhibit mitosis. The narrow-gap rheometer may be an efficient tool for drug screening and improving our understanding of mechanical issues in human disease treatment.

Cell stiffness under small and large deformations measured by optical tweezers and atomic force microscopy: effects of actin disruptors CK-869 and jasplakinolide

Spela Zemljic Jokhadar et al.

Summary: Cytoskeleton-disrupting drugs can have varied effects on cell mechanics depending on the deformation scales. The study found that actin-disrupting drugs CK-869 and jasplakinolide had different impacts on cellular stiffness, with CK-869 leading to decreased cell stiffness and jasplakinolide causing both stiffening and softening effects depending on the deformation scale. These results suggest that the effects of these drugs on cell mechanics are scale-dependent and vary based on the level of actin organization.

The epithelial-mesenchymal transition and the cytoskeleton in bioengineered systems

Susan E. Leggett et al.

Summary: The process of epithelial-mesenchymal transition (EMT) is intricately linked to alterations in the intracellular cytoskeleton and the extracellular matrix, leading to cellular elongation and polarity changes. Recent bioengineering approaches have shed light on the mechanisms behind EMT and cytoskeletal dynamics, with a focus on multicellular behavior, vimentin function, and contact guidance patterning. These technologies allow for a deeper understanding of EMT at the single cell level and offer insights into its implications for cancer progression.

Buffered λ-DNA solutions at high shear rates

H. Dakhil et al.

Summary: The study investigated buffered aqueous solutions of deoxyribonucleic acid isolated from bacteriophage lambda (lambda-DNA) at shear rates up to 10(5) s(-1) using a narrow-gap rheometer. The results showed that at high shear rates, viscosity levels off into an infinite-shear viscosity plateau. Additionally, normal-stress differences exhibited a power-law dependence on the shear rate close to previous findings at lower shear rates, but a stepwise change was observed beyond 10(4) s(-1) shear rates.

A Hydrogel Platform that Incorporates Laminin Isoforms for Efficient Presentation of Growth Factors - Neural Growth and Osteogenesis

Oana Dobre et al.

Summary: Laminins are crucial structural proteins in the ECM that bind growth factors and play tissue-specific roles. A new LM-PEG hydrogel platform inspired by Matrigel offers controlled stiffness and degradability, enabling specific targeting of tissues through GFs presented at ultralow doses. This platform demonstrates the potential to induce stem cell differentiation and promote neural cell growth in a synthetic, reproducible 3D microenvironment mimicking the native ECM.
