Quantification of Biologically and Chemically Bound Phosphorus inActivated Sludge from Full-Scale Plants with Biological P-Removal

Summary: Phosphorus exists in different forms in activated sludge, including metal salt precipitates, extracellular polymeric substances, and bound into biomass. This study combines various methods to obtain a comprehensive P mass-balance of activated sludge from EBPR plants. Chemically bound P constitutes the largest proportion of total P, while poly-P is found in bacterial cells.

The novel genus, 'Candidatus Phosphoribacter', previously identified as Tetrasphaera, is the dominant polyphosphate accumulating lineage in EBPR wastewater treatment plants worldwide

Summary: The bacterial genus Tetrasphaera is responsible for enhanced biological phosphorus removal (EBPR) in wastewater treatment plants. Recent analyses showed that 16S rRNA genes cannot accurately resolve the lineage of Tetrasphaera, which actually consists of several different genera within the Dermatophilaceae. This study identifies and examines 14 metagenome-assembled genomes containing full-length 16S rRNA genes from wastewater treatment plants, and reveals that the uncultured Tetrasphaera clade 3 represents two distinct genera named Ca. Phosphoribacter and Ca. Lutibacillus. Ca. Phosphoribacter is found to be the most abundant former Tetrasphaera lineage and polyphosphate accumulating organisms (PAO) in EBPR systems in Denmark and globally.

Reevaluation of the Phylogenetic Diversity and Global Distribution of the Genus Candidatus Accumulibacter

Summary: Candidatus Accumulibacter is an important polyphosphate-accumulating organism (PAO) in wastewater phosphorus removal. However, its taxonomic classification at the species level is complicated. This study provides a comprehensive reassessment of the phylogeny using genome, ppk1, and 16S rRNA gene-based approaches, resulting in the identification of novel species and clarification of the taxonomy. Additionally, the metabolic capabilities of Candidatus Accumulibacter species were explored using genome mining and in situ validation, providing insights into their niche differentiation and contribution to nutrient removal.

The transcriptionally most active populations included several PAOs, a competing glycogen accumulating organism, and other non-PAO microorganisms. The potential cooperation between these different populations may help reduce the accumulation of toxic nitric oxide.
