Biotechnological production of cyclic dinucleotides-Challenges and opportunities

Tabea Bartsch et al.

Summary: Cyclic dinucleotides (CDNs), as strong agonists of the stimulator of interferon genes, have great potential in pharmaceutical applications. The synthesis of CDNs through chemical and biocatalytic methods has its advantages and challenges. This review discusses the latest discoveries and trends in CDN synthesis, with a focus on the most promising biotechnological processes for CDN production.

Wonsuk Chang et al.

Summary: Stereochemically and structurally complex cyclic dinucleotide-based stimulator of interferon genes (STING) agonists were designed and synthesized, leading to the discovery of MK-1454 as a potential molecule for clinical development. It exhibited robust tumor cytokine upregulation and effective antitumor activity in mouse models, especially when used in combination with a fully murinized antimouse PD-1 antibody, mDX400.

Enzymatic Synthesis of 3′-5′, 3′-5′ Cyclic Dinucleotides, Their Binding Properties to the Stimulator of Interferon Genes Adaptor Protein, and Structure/Activity Correlations

Barbora Novotna et al.

Summary: This study examined the substrate specificity of four bacterial dinucleotide synthases and synthesized 24 3'3'CDNs. The prepared heterodimeric 3'3'CDNs showed better binding to STING and similar or better potency compared to bacterial 3'3'CDNs. Computational chemistry methods were used to analyze the interaction free energies between STING and CDNs, revealing promising correlations between computed and experimental results.

Antiviral Activity of 7-Substituted 7-Deazapurine Ribonucleosides, Monophosphate Prodrugs, and Triphoshates against Emerging RNA Viruses

Nemanja Milisavljevic et al.

Summary: A series of 7-deazaadenine ribonucleosides and their derivatives were synthesized and tested for antiviral activity against various RNA viruses. The modified triphosphates showed good antiviral activity by inhibiting viral RNA-dependent RNA polymerases, while nucleosides with bulkier heterocycles had lower toxicity.

Ligand Strain and Its Conformational Complexity Is a Major Factor in the Binding of Cyclic Dinucleotides to STING Protein

Miroslav Smola et al.

Summary: The study investigates the mechanism by which STING regulates immune responses, analyzing the factors contributing to the different affinities of CDNs ligands through structural and computational models. Experimental results suggest that the higher affinity of fluorinated analogues is due to smaller loss of entropy and strain energy upon binding.

Protein-Ligand Interactions in the STING Binding Site Probed by Rationally Designed Single-Point Mutations: Experiment and Theory

Zdenek Vavrina et al.

Summary: This study delves into the detailed examination of STING protein mutations interacting with six typical CDNs, showing that different mutations can either enhance or inhibit various types of protein-ligand interactions. The experimental data correlated well with theoretical calculations, providing a mechanistic interpretation of the differences in CDN binding to STING. This work may inspire the development of high-affinity ligands targeting key protein-ligand interactions against common STING haplotypes.

Synthesis and Biological Evaluation of Phosphoester and Phosphorothioate Prodrugs of STING Agonist 3′,3′-c-Di(2′F,2′dAMP)

Marketa Pimkova Polidarova et al.

Summary: In this study, the activity of 3',3'-c-di(2'F,2'dAMP) and its analogues against different STING allelic forms was evaluated. The synthesis of acyloxymethyl and isopropyloxycarbonyl phosphoester prodrugs of CDNs was described, and masking the negative charges of CDNs resulted in significantly improved activity of the prodrugs. Rapid uptake and intracellular cleavage of pivaloyloxymethyl prodrugs to the parent CDN were also observed, with peak intracellular concentration reached within 2 hours.
