Extracellular vesicles miR-210 as a potential biomarker for diagnosis and survival prediction of oral squamous cell carcinoma patients

Elisabetta Bigagli et al.

Summary: The expression of EV-miR-210 is significantly elevated in OSCC patients, showing potential as a diagnostic biomarker with high sensitivity and specificity. High EV-miR-210 expression is associated with worse 3-year survival, along with G2 and G3 grading and pN status being independent predictors of poor survival in OSCC patients.

The dual role of glioma exosomal microRNAs: glioma eliminates tumor suppressor miR-1298-5p via exosomes to promote immunosuppressive effects of MDSCs

Yanhua Qi et al.

Summary: Clear evidence shows that tumors can use exosomes to secrete microRNAs for modulating the tumor microenvironment. In this study, miRNAs were found to be divided into exosome-enriched miRNAs and intracellular-retained miRNAs, with exosome-enriched miRNAs playing a dual role. Specifically, miR-1298-5p was enriched in CSF exosomes and demonstrated to suppress glioma progression while promoting the immunosuppressive effects of myeloid-derived suppressor cells (MDSCs). Mechanically, miR-1298-5p targeted SETD7 and MSH2 in glioma cells and MDSCs, respectively. These findings provide insights into the selective sorting of miRNAs into exosomes and their role in modulating tumor immunity in glioma.

Risk stratification of postoperative recurrence in hypopharyngeal squamous-cell carcinoma patients with nodal metastasis

Hanqing Lin et al.

Summary: Risk factors such as pT3-T4, extranodal extension, lymphovascular invasion, and lower lymph node involvement were associated with poorer recurrence-free survival in HPSCC patients with nodal metastasis. Adjuvant chemoradiation significantly improved RFS in high-risk group patients, while low- and intermediate-risk group patients did not benefit significantly from adjuvant therapies in terms of RFS. The risk stratification based on these factors can effectively predict postoperative recurrence risk in N + HPSCC patients.

Selective exosome exclusion of miR-375 by glioma cells promotes glioma progression by activating the CTGF-EGFR pathway

Xiangdong Xu et al.

Summary: MiR-375 expression is downregulated in gliomas, and it suppresses glioma proliferation, migration, and invasion by inhibiting the CTGF-EGFR signaling pathway. MiR-375-containing exosomes were found in human blood samples from glioma patients, with levels correlating with disease progression. Exosomal miR-375 secretion impacts the activity of the CTGF-EGFR pathway, and once secreted, exosomal miR-375 is not taken back up by glioma cells.

Assessment and treatment strategies for occult contralateral lymph node metastasis in hypopharyngeal squamous cell carcinoma patients with ipsilateral node-positive necks

Yu Heng et al.

Summary: This study aims to quantitatively predict the risk of occult contralateral lymph node metastasis in HSCC patients with ipsilateral lymph node metastasis, and construct two nomograms to guide treatment selection. Multivariate analyses identified several factors as independent risk factors for contralateral lymph node metastasis, and the nomograms provide guidance on the necessity of contralateral lymph node dissection and therapeutic radiation targeting. High-risk patients should receive contralateral lymph node dissection, while moderate-risk and low-risk patients require varying levels of follow-up and treatment.

A methyltransferase-like 14/miR-99a-5p/tribble 2 positive feedback circuit promotes cancer stem cell persistence and radioresistance via histone deacetylase 2-mediated epigenetic modulation in esophageal squamous cell carcinoma

Zhenchuan Liu et al.

Summary: The study identified a positive feedback loop involving METTL14/miR-99a-5p/TRIB2 that enhances CSC characteristics and radioresistance in ESCC. It was found that miR-99a-5p inhibits CSC persistence and radioresistance, while TRIB2 activates HDAC2 leading to epigenetic repression of p21 through the Akt/mTOR/S6K1 signaling pathway. Targeting this axis may provide potential therapeutic targets for ESCC treatment.

MicroRNA-99a-5p suppresses cell proliferation, migration, and invasion by targeting isoprenylcysteine carboxylmethyltransferase in oral squamous cell carcinoma

Xiang Sun et al.

Summary: MiR-99a-5p expression is significantly decreased in oral squamous cell carcinoma (OSCC) tissues and cell lines, and is associated with clinical stage and lymph node metastasis in OSCC. Overexpression of miR-99a-5p inhibits proliferation, migration, and invasion abilities of OSCC cells by directly targeting ICMT. These results suggest that miR-99a-5p could serve as a potential therapeutic target for OSCC treatment.
