Pathways to cures for multiple sclerosis: A research roadmap

Bruce F. Bebo et al.

Summary: This article presents a research roadmap for curing multiple sclerosis (MS) and provides recommendations for accelerating the development of MS cures. The roadmap was developed through the engagement of scientific thought leaders and individuals affected by MS, and it was endorsed by leading MS organizations. The roadmap consists of three distinct but overlapping cure pathways, including stopping the MS disease process, restoring lost function, and ending MS through prevention.

Genetic disorders with central nervous system white matter abnormalities: An update

Anju Shukla et al.

Summary: This article reviews 422 genetic conditions with white matter abnormalities, covering definitions, classification, clinical spectrum, neuroimaging findings, genomics, and molecular mechanisms. While diagnosis relies on genomic tests, clinical and neuroimaging findings remain relevant. The current understanding of pathophysiology and therapeutics of these disorders is also discussed.

The Current Challenges for Drug Discovery in CNS Remyelination

Sonia Balestri et al.

Summary: The myelin sheath wraps around axons for saltatory currents transmission and can be damaged by various factors, leading to neurodegeneration. Drugs promoting CNS remyelination are promising but face challenges, such as timing of administration and failure in neurodegenerative pathologies, as well as the need to recreate disease-relevant environments for drug screening. Recent biological findings on oligodendrocyte maturation and brain repair post-myelin injury may impact future drug screening campaigns for CNS regenerative drugs.
