trans-4-Fluoro-L-proline: A Sensitive 19F NMR Probe for the Rapid Simultaneous Enantiomeric Analysis of Multicomponent Amines

Summary: Simultaneous enantiomeric analysis is crucial for medicine, food security, and life science. This study introduces a novel chiral derivatizing agent, CDApro, based on trans-4-fluoro-L-proline for rapid and simultaneous analysis of various chiral amine mixtures, including non-amino acid amines, multi-component amino acids, and amines with chiral centers several carbons remote to the amino group. The CDApro reagent serves as a robust and powerful tool for determination of enantiomeric purity and quantification of chiral amines.

Electrostatic effects on 19F NMR chemical shifts in N-phenyl g-lactam derivatives

Summary: Covalently identical ortho-F nuclei display dual 19F NMR signals in di-ortho-F substituted N-phenyl glactams. Structural variations at C9-substituent on the g-lactam lead to larger chemical shift changes at downfield 19F signals (ddown) than those at upfield 19F signals (dup). A reverse correlation between downfield 19F chemical shifts and electron-withdrawing abilities of C9-phenylamides is observed. Electrostatic effects are the major forces between C9-substituents and ortho-F nuclei, and Hammett parameters can be used to describe such through-space interactions. Structural factors influencing ortho-19F shielding among diverse C9-substituted N-phenyl g-lactams are discussed.

Tailoring Sensors and Solvents for Optimal Analysis of Complex Mixtures Via Discriminative 19F NMR Chemosensing

Summary: The study systematically investigated the influence of sensor structure and NMR solvents on the resolution of structurally similar analytes, finding that substituents adjacent and distal to the F-19 labels are both important to the resolving ability of the F-19-labeled sensors. More pronounced separation between F-19 NMR peaks was observed in nonpolar and aromatic solvents, and using a proper sensor and solvent combination, more than 20 biologically relevant analytes can be simultaneously identified.
