Bioorthogonal click and release: A general, rapid, chemically revertible bioconjugation strategy employing enamine N-oxides

Enamine N-Oxides: Synthesis and Application to Hypoxia-Responsive Prodrugs and Imaging Agents

Summary: Tumor hypoxia-induced adaptive reprogramming of cancer cells can lead to poor prognosis, making hypoxia-activated prodrug development challenging. A novel hypoxia-responsive trigger derived from enamine N-oxide structures was synthesized and applied for hypoxia-activated prodrug and tumor labeling. The versatile enamine N-oxide motif with dual drug release and imaging modalities presents an attractive opportunity for theranostic development to address the needs for new therapeutics and patient stratification.
