Engineering of Aeromonas caviae Polyhydroxyalkanoate Synthase Through Site-Directed Mutagenesis for Enhanced Polymerization of the 3-Hydroxyhexanoate Unit

Ken Harada et al.

Summary: In this study, the PhaC(Ac) enzyme was engineered to increase the 3HHx fraction in biosynthesized PHA. By targeting specific residues based on three-dimensional structural information, a triple mutant with increased 3HHx fraction was successfully engineered for the biosynthesis of a higher 3HHx-fraction polymer.

Summary: PHBHHx is a practical bacterial polyhydroxyalkanoate produced by recombinant strains of Cupriavidus necator from CO2. The composition of PHBHHx can be controlled by using different enzymes, and its synthesis can be achieved using a mixture of H-2/O-2/CO2 and NH4)2SO4 as a nitrogen source.
