A Useful Guide to Lectin Binding: Machine-Learning Directed Annotation of 57 Unique Lectin Specificities

Daniel Bojar et al.

Summary: Glycans play critical roles in biology and medicine, but the specificity of lectins, which are key proteins in glycans binding, has not been well-defined. In this study, machine learning algorithms and expert annotation were used to define the lectin specificity for 57 unique lectins. This research provides important insights into the complex binding features of commercially available lectins.

Sialic acid-containing glycolipids mediate binding and viral entry of SARS-CoV-2

Linh Nguyen et al.

Summary: Evidence suggests that host glycans, specifically glycolipids containing sialic acid, play a role in facilitating the entry of SARS-CoV-2 virus into host cells by binding to the receptor-binding domain (RBD) of the spike protein. Depletion of cell surface sialic acid levels through various methods decreases RBD binding and infection of the virus, indicating the importance of sialylated glycans in viral entry.

Synthetic Heparan Sulfate Mimetic Pixatimod (PG545) Potently Inhibits SARS-CoV-2 by Disrupting the Spike-ACE2 Interaction

Scott E. Guimond et al.

Summary: This study found that the synthetic HS mimetic pixatimod can interact with the receptor binding domain of the SARS-CoV-2 spike protein and directly inhibit its binding to ACE2. The results show that pixatimod can effectively inhibit the infection of multiple clinical isolates of SARS-CoV-2 virus and has broad potency against various variants. In a mouse model, pixatimod also reduces viral titers and weight loss. This study provides strong evidence for the potential use of synthetic HS mimetics as a therapeutic approach for COVID-19.

Glycosylated gold nanoparticles in point of care diagnostics: from aggregation to lateral flow

Alexander N. Baker et al.

Summary: This article introduces the potential of glycans coupled to gold nanoparticles as recognition agents for lateral flow diagnostics. The concept of lateral flow is first introduced, with a case study comparing its use in the field to other diagnostic tools. The use of glycosylated materials and the affinity gains achieved by the cluster glycoside effect are then discussed. Finally, the potential role of glycans in lateral flow is explained, and examples of their successful use are provided.
