FALC suppresses cell growth and induces apoptotic cell death, while also activating autophagy and inhibiting metastasis. These findings suggest that FALC may be a novel candidate for therapeutic strategy against OSCCs.

E3 ligase RNF167 and deubiquitinase STAMBPL1 modulate mTOR and cancer progression

Summary: This study identifies RNF167 and STAMBPL1 as key regulators of Sestrin2 polyubiquitination in response to leucine availability. The decreased expression of RNF167 and increased expression of STAMBPL1 are found in gastric and colorectal tumors. Knockout of STAMBPL1 or correction of the STAMBPL1 mutation suppresses xenograft tumor growth in a human colon cancer cell line. Furthermore, a cell-permeable peptide that blocks STAMBPL1-Sestrin2 interaction inhibits mTORC1 and could be a potential option for cancer therapy.

Summary: miR-18b-5p is downregulated while PTEN is upregulated in follicular fluid exosomes of PCOS patients. Decreasing PTEN promotes KGN cell proliferation and limits apoptosis. Exosomes stimulate KGN cell proliferation and suppress apoptosis, decrease FSH, LH, and testosterone levels, and increase E2 levels. Upregulating miR-18b-5p enhances the inhibitory effects of exosomes on PCOS progression. miR-18b-5p improves PCOS by targeting PTEN and activating the PI3K/Akt/mTOR pathway. Circulating miR-18b-5p levels are associated with the progression of PCOS complications.

Summary: Mechanistic target of rapamycin (mTOR) is a protein kinase that regulates cell functions. Dysregulation of mTOR activity is associated with various pathological conditions. Inhibition of overactivated mTOR is a rational approach for treating human diseases. Rapamycin is a natural inhibitor of mTOR with antitumor and immunosuppressive activity. Different generations of mTOR inhibitors have been developed, including rapalogs, mTOR kinase inhibitors, and dual PI3K/mTOR inhibitors. Novel inhibitors are still being developed to better understand the role of mTOR in signaling pathways and human diseases.
