Effect of Chitosan Deacetylation on Its Affinity to Type III Collagen: A Molecular Dynamics Study

Piotr Beldowski et al.

Summary: The ability of linear glucosamine polysaccharides to form strong intermolecular interactions with collagen is closely related to their nonlinear dynamic behavior and bio-lubricating features. This study found a linear relationship between the degree of deacetylation and the collagen-chitosan interaction energy, with increasing deacetylation degree resulting in stronger interaction. This can be explained by the replacement of weak hydrophobic contacts with more stable hydrogen bonds involving amino groups in N-deacetylated chitosan. The affinity of chitosan for collagen was also found to be higher than that of hyaluronic acid when the degree of deacetylation was greater than 0.4.

Prokaryotic Collagen-Like Proteins as Novel Biomaterials

Jonathan Picker et al.

Summary: Collagens, as the major structural component in animal extracellular matrices, play crucial roles in cell-matrix interactions. The unique triple helical structure of collagens is enabled by Gly-X-Y repeats. Prokaryotic collagen-like proteins, which were discovered in the early 2000s, have similar structures to mammalian collagens and can be easily expressed in E. coli and genetically modified. This review article discusses the properties of mammalian collagen, compares mammalian collagen with prokaryotic collagen-like proteins, and reviews the applications of prokaryotic collagen-like proteins in studying collagen biology and developing new biomaterial platforms. Furthermore, it describes the application of the streptococcal collagen-like protein Scl2 in thromboresistant coating, scaffolds for bone regeneration, chronic wound dressing, and matrices for cartilage regeneration.

Contrasting Local and Macroscopic Effects of Collagen Hydroxylation

Sameer Varma et al.

Summary: The study found that removal of hydroxylation has minimal effect on the macroscopic structure of collagen, but does have some impact on local structure, hydrogen bonding capacity, and electrostatic attraction. Experimental observations show that hydroxylation removal has minimal effect on collagen's D-band length, gap-overlap ratio, monomer width, and monomer length. Interestingly, de-hydroxylation also has a minor effect on the fibril's Young's modulus, with accompanying changes in triple-helix windings.
