Mapping genomic loci implicates genes and synaptic biology in schizophrenia

Vassily Trubetskoy et al.

Summary: In this study, a two-stage genome-wide association study was conducted to identify common variants associated with schizophrenia. The results revealed 287 distinct genomic loci and 120 genes likely to be involved in the development of schizophrenia. This research provides valuable insights into the pathophysiology of schizophrenia and offers a resource for further mechanistic studies.

Exome sequencing in bipolar disorder identifies AKAP11 as a risk gene shared with schizophrenia

Duncan S. Palmer et al.

Summary: Exome sequencing analysis of individuals with bipolar disorder reveals enrichment of ultra-rare protein-truncating variants in constrained genes. AKAP11 is identified as a risk gene for both bipolar disorder and schizophrenia. This study highlights the importance of rare coding variation in the development of bipolar disorder.

Genome-wide analysis of schizophrenia and multiple sclerosis identifies shared genomic loci with mixed direction of effects

Mohammad Ahangari et al.

Summary: Despite a negligible genetic correlation, there is polygenic overlap between schizophrenia and multiple sclerosis, with substantial genetic enrichment in each disorder conditional on associations with the other. Leveraging this cross-disorder enrichment, 36 loci jointly associated with schizophrenia and multiple sclerosis were identified at conjunctional FDR < 0.05 with mixed direction of effects. By conducting functional analysis, candidate genes and biological processes related to immune response and B-cell receptor signaling pathways were implicated in the shared genetic architecture of these two disorders.

Bipolar multiplex families have an increased burden of common risk variants for psychiatric disorders

Till F. M. Andlauer et al.

Summary: This study analyzed whether the risk for bipolar disorder in BD multiplex families is influenced by common genetic variants. The results showed that familial BD cases and unaffected family members had higher PRS for all three psychiatric disorders, indicating a high baseline risk in the families; familial BD cases had significantly higher BD PRS than unaffected family members and unrelated BD cases, suggesting that development of BD in these families is attributable to common variants with specific risk for BD.

Pleiotropy and Cross-Disorder Genetics Among Psychiatric Disorders

Phil H. Lee et al.

Summary: Genome-wide analyses have shown the important role and widespread nature of pleiotropy in the genetic basis of psychiatric disorders, with genetic influences transcending clinical diagnostic boundaries. The observed pleiotropy extends from overall genome-wide correlations to biological pathways and genetic effects at individual loci.

Investigating Shared Genetic Basis Across Tourette Syndrome and Comorbid Neurodevelopmental Disorders Along the Impulsivity-Compulsivity Spectrum

Zhiyu Yang et al.

Summary: The study investigates the shared genetic basis across TS, ADHD, ASD, and OCD, revealing a common unifying factor connecting TS, ADHD, and ASD, while showing the highest genetic correlation between TS and OCD. Through a systematic meta-analysis, multiple novel hits and regions have been identified, indicating pleiotropic roles for specific disorders analyzed here. The study underlines the importance of redefining the framework for research across traditional diagnostic categories.

Investigating the genetic architecture of noncognitive skills using GWAS-by-subtraction

Perline A. Demange et al.

Summary: Genomic structural equation modeling was used to estimate the genetic component of educational attainment independent of cognitive ability, finding that 57% of genetic variation in educational attainment is attributed to noncognitive skills. The study identified significant loci and a polygenic architecture contributing to educational attainment, with noncognitive genetics showing associations with personality traits, behavior, and psychiatric disorders. Noncognitive and cognitive-performance genetics were found to be similarly associated with socioeconomic success and longevity.
