Human organoid biofilm model for assessing antibiofilm activity of novel agents

Summary: A human epidermis organoid model was developed for studying host-microbe interplay and screening for novel antibiofilm agents. Treatment with antibiofilm peptide DJK-5 significantly reduced bacterial burden of MRSA and PAO1 biofilms. Application of DJK-5 on thermally wounded skin infected with MRSA biofilms led to decreased bacterial load, cytotoxicity, and pro-inflammatory cytokine levels.

Virulence Gene Expression of Staphylococcus aureus in Human Skin

Summary: The study established a cutaneous infection model using human skin explants to investigate the gene expression profile of virulence-related factors of S. aureus USA300 LAC strain. 12 virulence factors were identified as consistently upregulated in skin tissues at all time points assessed. Confocal microscopy revealed that the expression of alpha-hemolysin by S. aureus varied dependent on the skin niche and bacteria preferentially accumulated inside sweat glands and ducts, providing insights for the development of therapeutic strategies against S. aureus.
