Utilization of Treg Cells in Solid Organ Transplantation

Tanya Juneja et al.

Summary: Organ transplants are a life-saving treatment for patients with end-stage organ failure. However, the long-term outcomes for these patients are poor due to the use of immunosuppressants, which increase the risk of infections and damage. Regulatory T cells (Tregs) are being investigated as a potential treatment to minimize rejection and prevent negative outcomes.

Tracking Regulatory T Cell Development in the Thymus Using Single-Cell RNA Sequencing/TCR Sequencing

David L. Owen et al.

Summary: Recent studies have revealed multiple stages of regulatory T cell development in the thymus, with both CD25+ T cell progenitors and FOXP3lo T cell progenitors undergoing agonist-activated states and transitional stages before differentiating into mature T cells. CD25+ T cell progenitors were found to be the major source of IL-2 required for the conversion of progenitors into mature T cells. Additionally, thymic resident regulatory T cells were found to be heterogeneous and exhibit the most diverse T cell receptor repertoire among CD4 single-positive thymocytes.

Summary: This study reevaluated the role of IL-2 and IL-15 in the development of regulatory T-lymphocytes (Treg) and found that IL-2 and IL-15 play important non-redundant quantitative and qualitative roles in intrathymic Treg development.

BATF epigenetically and transcriptionally controls the activation program of regulatory T cells in human tumors

Kota Itahashi et al.

Summary: Research has shown that Treg cells in the TME undergo a distinct open chromatin profile, with key transcription factors including BATF, IRF4, NF-κB, and NR4A, among which BATF serves as a crucial regulator influencing the differentiation and activation of Treg cells in the TME through epigenetic control of activation-associated gene expression. BATF deficiency may inhibit tumor growth, and high levels of BATF expression are associated with poor prognosis.

Recirculating IL-1R2+ Tregs fine-tune intrathymic Treg development under inflammatory conditions

Eirini Nikolouli et al.

Summary: A specific subset of Foxp3(+) Tregs expressing IL-1R2 has been identified in the thymus, showing characteristics of recirculating cells with an activated phenotype. These Tregs have a thymic origin and play a regulatory role in modulating IL-1 signaling in the thymus.

Specialized regulatory T cells control venous blood clot resolution through SPARC

Fatemeh Shahneh et al.

Summary: The cells and mechanisms involved in blood clot resorption are only partially known, with regulatory T cells playing a crucial role in regulating thrombolysis by controlling monocytes. The matricellular protein SPARC is shown to enhance monocyte activity and is crucial for blood clot resorption. Expanding Tregs within a therapeutic window can accelerate clot resorption.

Kidney GATA3+ regulatory T cells play roles in the convalescence stage after antibody-mediated renal injury

Ryota Sakai et al.

Summary: Tregs accumulation in the kidney is crucial for disease control, with GATA3 playing an important role in recruiting Tregs to the kidney. The experimental data suggest that PPAR gamma agonist enhances the accumulation of GATA3(+) Tregs in the kidney.

T reg cell-intrinsic requirements for ST2 signaling in health and neuroinflammation

Saskia Hemmers et al.

Summary: The study suggests that ST2 expression on regulatory T cells is not essential for their accumulation and residence in nonlymphoid organs, but cell-intrinsic sensing of IL-33 contributes to cytokine production in certain tissues. Additionally, regulatory T cells play a crucial role in limiting IL-17A-producing gammadelta T cells in the central nervous system.

Single-cell chromatin accessibility landscape identifies tissue repair program in human regulatory T cells

Summary: Research identified features of human tissue Treg cells with tissue repair functions, which show similarities in gene expression profiles at the single-cell level with murine cells, and shared characteristics with T follicular helper-like cells. Inducing differentiation of human Treg cells into Tfh-like cells can partially replicate the regenerative characteristics of tissue Treg cells.

Interleukin-33-Dependent Accumulation of Regulatory T Cells Mediates Pulmonary Epithelial Regeneration During Acute Respiratory Distress Syndrome

Summary: Research has shown that the absence of Treg cells in ARDS can result in impaired lung repair, with IL-33 signaling playing a crucial role in the accumulation of Treg cells. Moreover, promoting the regeneration of Treg cells through the transfer of exogenous IL-33 can facilitate pulmonary epithelial regeneration in ARDS.
