Activation of the intestinal tissue renin-angiotensin system by transient sodium loading in salt-sensitive rats

Masaki Ryuzaki et al.

Summary: This study reveals the previously unknown contribution of the intestinal tissue renin-angiotensin system to sodium homeostasis and blood pressure regulation in the pathophysiology of salt-sensitive hypertension. Transient salt loading in salt-sensitive rats leads to sustained blood pressure elevation, decreased fecal sodium content, and excess circulating volume. Administration of an angiotensin II type 1 receptor blocker can effectively reduce blood pressure and normalize fecal sodium levels. Furthermore, transient salt loading induces the intestinal tissue renin-angiotensin system and enhances sodium transporter expression exclusively in the small intestine of salt-sensitive rats.

GLP-1 Mediates Regulation of Colonic ACE2 Expression by the Bile Acid Receptor GPBAR1 in Inflammation

Michele Biagioli et al.

Summary: This study investigates the regulation of ACE2 expression in Crohn's disease patients and rodent models of colitis. The results suggest that ACE2 expression is regulated by TNF-α and GLP-1, and GPBAR1 acts as a positive modulator of ACE2.

Perindopril/Ambrosin Combination Mitigates Dextran Sulfate Sodium-Induced Colitis in Mice: Crosstalk between Toll-Like Receptor 4, the Pro-Inflammatory Pathways, and SIRT1/PPAR-γ Signaling

Ahmed M. Kabel et al.

Summary: This study investigated the effects of perindopril and ambrosin, alone or in combination, on DSS-induced colitis in mice. The results showed that both perindopril and ambrosin were able to alleviate oxidative stress, suppress proinflammatory pathways, inhibit apoptosis, and enhance autophagy in colonic tissues. The combination of perindopril and ambrosin had the most remarkable effects compared to either agent alone.

Intestinal Inflammation Modulates the Expression of ACE2 and TMPRSS2 and Potentially Overlaps With the Pathogenesis of SARS-CoV-2-related Disease

Summary: The study shows that the COVID-19 virus has the ability to invade the intestines, and medications used by IBD patients do not significantly affect receptor expression in uninflamed intestines. There are shared molecular networks between COVID-19 infection and IBD.

Intestinal Receptor of SARS-CoV-2 in Inflamed IBD Tissue Seems Downregulated by HNF4A in Ileum and Upregulated by Interferon Regulating Factors in Colon

Bram Verstockt et al.

Summary: In patients with inflammatory bowel disease, intestinal inflammation alters the coreceptors for SARS-CoV-2 in the intestine, leading to dysregulation of ACE2 in the ileum and colon. Inflammatory stimuli upregulate ACE2 expression in organoids from UC patients, while anti-TNF therapy restores proper ACE2 regulation in responders.

Soluble angiotensin-converting enzyme 2 is transiently elevated in COVID-19 and correlates with specific inflammatory and endothelial markers

Annika Lundstrom et al.

Summary: The study found that levels of sACE2 were higher in COVID-19 patients compared to healthy controls, but decreased over time while remaining higher than in controls. The correlations of sACE2 and sACE levels with markers of inflammation and endothelial dysfunction were different, suggesting release from different cell types and/or vascular beds.
