Summary: The evolution of tumor hallmarks is a result of tumor cells accommodating to their surrounding tumor microenvironment (TME), which is crucial for cell survival. Cancer cell metabolism depends significantly on the stroma, and hypoxia is a common event in solid cancers and a key hallmark of chronic inflammation. The production of Transforming Growth Factor (TGF)-beta in chronic inflammatory and hypoxic settings is considered a cardinal factor in inducing tumor hallmarks. Aging, obesity, and smoking are main predisposing factors for cancer, primarily acting through modulation of the TME.

Summary: Ovarian cancer is a highly aggressive disease, often diagnosed late after it has spread. Inflammatory signals and cellular senescence play important roles in ovarian cancer dissemination. Recent studies have shown that platinum-based or PARP inhibitor therapies, standard treatments for ovarian cancer, can induce senescence in ovarian cancer cells, leading to an inflammatory response known as the senescence-associated secretory phenotype (SASP). The SASP may promote metastasis in ovarian cancer.

Tumor-derived exosomes in hypoxic microenvironment: release mechanism, biological function and clinical application

Summary: Hypoxia, a characteristic of solid tumors, is closely related to the aggressiveness and unfavorable outcomes of diseases. Communication between cancer cells and their microenvironment plays a critical role in cancer progression, and recent studies have shown that this communication includes not only secreted molecules, but also exosomes secreted by tumor cells. Exosomes, nano-scale extracellular vesicles, carry molecular characteristics and cargo of the source cell, participating in intercellular communication through different pathways. Hypoxia has been found to influence the secretion and function of exosomes in various types of cancer. This review summarizes the physiological mechanism of hypoxia regulating exosome secretion, and discusses the involvement of hypoxic exosomes in cancer progression and immune escape processes, as well as the potential for targeting hypoxia-induced exosomes in cancer therapy strategies.

Summary: This study found that gastric cancer-derived sEVs are primarily absorbed by Kupffer cells, leading to high expression of sEV-miR-151a-3p in GC-LM patients with poor prognosis. These sEV-miR-151a-3p activate the SMAD2/3 pathway by inhibiting SP3 and targeting YTHDF3 in Kupffer cells, promoting liver metastasis of gastric cancer cells and enhancing stem cell-like properties. Additionally, sEV-miR-151a-3p induces miR-151a-3p transcription in Kupffer cells, forming a positive feedback loop to establish a hepatic stemness-permissive niche to support GC-LM.
