ICAM-1-mediated adhesion is a prerequisite for exosome-induced T cell suppression

Wei Zhang et al.

Summary: This study reveals the co-localization of ICAM-1 and PD-L1 on exosomes, both of which are upregulated by interferon-y. The adhesion between TEVs and T cells mediated by ICAM-1-LFA-1 is crucial for exosomal PD-L1-mediated immune suppression.

Primary tumor associated macrophages activate programs of invasion and dormancy in disseminating tumor cells

Lucia Borriello et al.

Summary: This study utilizes high-resolution intravital imaging to track breast-derived metastatic cells in the lung and finds that macrophages in the primary tumor affect the dissemination and dormancy phenotypes of these cells.

Ral GTPases promote breast cancer metastasis by controlling biogenesis and organ targeting of exosomes

Shima Ghoroghi et al.

Summary: This study identifies Ral GTPases as central molecules linking the mechanisms of EV secretion and cargo loading to their capacity to disseminate and induce pre-metastatic niches. Depletion of RalA or RalB in cells leads to limited organotropic capacities in vivo and less efficient promotion of metastasis, as well as reduced levels of the adhesion molecule MCAM/CD146 in EVs, which favors EV-mediated metastasis by targeting to the lungs. RalA, RalB, and MCAM/CD146 are factors of poor prognosis in breast cancer patients.

The cancer glycocalyx mediates intravascular adhesion and extravasation during metastatic dissemination

Giovanni S. Offeddu et al.

Summary: Offeddu et al. used a microfluidic vascular model to study the effects of selectively removing glycocalyx components from cancer cells. They found that cancer-associated hyaluronic acid and CD44 mediate firm adhesion to endothelial cells and extravasation during metastasis.

Metastasis-Initiating Cells and Ecosystems

Joan Massague et al.

Summary: Metastasis-initiating cells (MIC) are cancer cells with stem-like and immune-evasive properties that adopt regenerative progenitor phenotypes, enabling them to survive the stresses of metastasis and evade immune surveillance. Understanding the molecular mechanisms of MIC phenotypes and host tissue ecosystems is crucial for developing improved cancer therapeutics.

ICAM1 initiates CTC cluster formation and trans-endothelial migration in lung metastasis of breast cancer

Tumor extracellular vesicles drive metastasis (it's a long way from home)

Shima Ghoroghi et al.

Summary: Extracellular vesicles released by tumors play a crucial role in the process of metastasis by inducing the formation of premetastatic niches, creating a favorable microenvironment for circulating tumor cells to seed and colonize. Tumor EV cargos have significant effects on various biological processes, such as extracellular matrix remodeling and angiogenesis.
