Twisting DNA by salt

Sergio Cruz-Leon et al.

Summary: The structure and properties of DNA are influenced by the environment, particularly the ion atmosphere. This study investigates how DNA twist changes with the concentration and identity of surrounding ions. The results show that DNA twist increases with increasing concentration for all ions studied, and the twist strongly depends on the specific ion identity. Molecular dynamics simulations provide a microscopic explanation for the observed ion specificity.

Co-condensation of proteins with single- and double-stranded DNA

Roman Renger et al.

Summary: This study investigated the mechanism of how the FUS protein forms liquid-like assemblies by co-condensing with DNA molecules in vitro. It was found that the protein could adsorb onto DNA to generate a sticky FUS-DNA polymer, which then collapsed to form a liquid-like FUS-DNA co-condensate. The co-condensation mechanism mediated by protein adsorption on nucleic acids is suggested to be important for intracellular compartmentalization.

DNA sequence-directed cooperation between nucleoid-associated proteins

Summary: This study investigated the formation of long-range nucleoprotein structures by two major NAPs, FIS and H-NS, binding to DNA molecules with distinct binding site arrangements using atomic force microscopy and solid-state nanopores. The spatial organization of protein binding sites was found to govern the higher-order architecture of the nucleoprotein complexes, highlighting the role of DNA sequence in driving structural differentiation within the bacterial chromosome.

DNA-loop-extruding SMC complexes can traverse one another in vivo

Hugo B. Brandao et al.

Summary: Hi-C analyses and 3D polymer simulations on engineered Bacillus subtilis strains reveal that SMC complex encounters on the same DNA are resolved via bypassing in vivo. Over 20 engineered strains show a variety of chromosome folding patterns, which require SMC complexes to bypass each other in vivo. This bypassing activity enables SMC complexes to avoid traffic jams while spatially organizing the genome.
