Self-Assembling Lectin Nano-Block Oligomers Enhance Binding Avidity to Glycans

Shin Irumagawa et al.

Summary: Lectins are proteins that have the ability to bind carbohydrates and they have multiple carbohydrate recognition domains and multivalent binding effects. This study developed functional protein nano-building blocks by fusing lectins to oligomeric proteins, which can self-assemble into various nanostructures. The binding avidity of the lectin nano-blocks to glycans was significantly increased through multivalent effects.

Strategies and Tactics for the Development of Selective Glycan-Binding Proteins

Elizabeth M. Ward et al.

Summary: Glycans have significant impacts on biological processes, disease states, and pathogenic interactions. Glycan-binding proteins, such as lectins, are crucial tools for investigating glycan structure and function, but the lack of specific and selective protein reagents hinders glycans research. Efforts have been made to expand the toolbox of GBPs beyond natural sources through protein engineering to enhance their properties and broaden their potential applications.

The Two Sweet Sides of Janus Lectin Drive Crosslinking of Liposomes to Cancer Cells and Material Uptake

Lina Siukstaite et al.

Summary: A chimeric, bispecific Janus lectin has been engineered with different recognition sites for simultaneous binding to sialylated and fucosylated glycoconjugates. This lectin shows nanomolar avidities for sialic acid and fucose, and has the potential for targeted drug delivery and tumor recognition. It has been demonstrated to mediate the internalization of liposomal content in human epithelial cancer cells, highlighting its utility in specific drug delivery applications.

Segregated Protein-Cucurbit[7]uril Crystalline Architectures via Modulatory Peptide Tectons

Kiefer O. Ramberg et al.

Summary: This study describes the modulation of protein assemblies mediated by cucurbituril through the inclusion of peptide tectons. By fusing different peptides to RSL, the researchers were able to create layered materials with varying porosity, showcasing the modularity of protein-cucurbituril architectures. This approach adds a new dimension to macrocycle-mediated protein assembly and could potentially lead to the development of new types of materials with segregated protein crystals.
