Generation and Screening of Antigen-Specific Nanobodies from Mammalian Cells Expressing the BCR Repertoire Library Using Droplet-Based Microfluidics

Menghua Lyu et al.

Summary: Nanobodies, derived from the serum of Camelidae, offer advantages over conventional antibodies and hold promise for immunotherapy and antibody-drug development. A high-throughput nanobody screening platform is crucial for their rapid development. While droplet-based microfluidic systems have shown improved performance compared to traditional phage display technology, directly applying this technology to nanobody screening faces challenges. This study overcame these obstacles by constructing a eukaryotic display system and designing a signal capture system, resulting in successful screening of target cells expressing anti-PD-L1 nanobodies.

Summary: Nanobodies, derived from Camelidae serum, have advantages over conventional antibodies and hold promise for immunotherapy and antibody-drug development. A high-throughput nanobody screening platform is crucial. This study developed a eukaryotic display system for nanobody screening and used single-cell transcriptomic profiling to uncover intercellular heterogeneity.

Unlocking the potential of antibody-drug conjugates for cancer therapy

Joshua Z. Drago et al.

Summary: Nine ADCs are currently approved for cancer treatment, with many more in development. Advances in synthetic biochemistry have led to a new generation of ADCs promising improved tissue specificity and cytotoxicity. Despite impressive activity against treatment-refractory cancers, challenges such as systemic toxicity and acquired resistance remain. This review discusses the design, mechanism of action, and limitations of ADCs, proposing strategies to maximize their therapeutic benefit for cancer patients.

Bacteriophage uptake by mammalian cell layers represents a potential sink that may impact phage therapy

Marion C. Bichet et al.

Summary: It has been found that bacteriophages have tropisms beyond their bacterial hosts and can adhere to mammalian tissues before being internalized through processes like macropinocytosis. Understanding these interactions may have significant implications on immune responses, phage pharmacokinetics, and the efficacy of phage therapy.
