Surfactant Treatment for Efficient Gene Detection of Enteric Viruses and Indicators in Surface Water Concentrated by Ultrafiltration

Summary: The HFUF-based microbial concentration method is commonly used for monitoring pathogenic viruses and microbial indicators in water samples, but it can co-concentrate substances that interfere with molecular processes. This study evaluated the efficacy of sodium deoxycholate (SD) treatment in mitigating these inhibitory substances. The SD treatment significantly improved the RNA extraction-RT-quantitative PCR efficiency and detection rates of indigenous viruses.

Application of Capsid Integrity (RT-)qPCR to Assessing Occurrence of Intact Viruses in Surface Water and Tap Water in Japan

Summary: The study applied a new method (SD-CDDP-(RT-)qPCR) to detect viruses in surface water and tap water in the Kanto region of Japan. The results showed that PMMoV was more frequently detected in surface water, while most target viruses were not detected in tap water. SD-CDDP-(RT-)qPCR revealed a higher proportion of intact PMMoV and a lower proportion of intact human pathogenic viruses.
