Paracellular permeability and tight junction regulation in gut health and disease

Arie Horowitz et al.

Summary: Increased intestinal permeability due to tight junction barrier loss can be targeted in gastrointestinal diseases. The authors discuss the molecular components and regulation of tight junctions, and their relevance to gut diseases and therapeutic opportunities. The restoration of increased unrestricted pathway permeability requires mucosal healing, while tight junction barrier loss can be reversed by targeted interventions. Specific approaches are needed to restore pore pathway or leak pathway permeability increases, and recent studies have demonstrated the potential of barrier restoration in disease.

A medium composition containing normal resting glucose that supports differentiation of primary human airway cells

Rachel Morgan et al.

Summary: This study successfully differentiated human basal cells isolated from the nasal and conducting airway into mature epithelial cells at the air-liquid interface (ALI) using a medium with normal resting glucose levels, avoiding the need for high glucose concentrations. The use of normal glucose-containing ALI medium also enabled differentiation of nasal and tracheal cells and allowed for long-term cell responsiveness.

Loss of GM-CSF-dependent instruction of alveolar macrophages in COVID-19 provides a rationale for inhaled GM-CSF treatment

Cedric Bosteels et al.

Summary: GM-CSF plays a crucial role in COVID-19, but excessive use may lead to inflammation. Inhalation of GM-CSF does not improve oxygenation parameters, but it enhances antiviral immune response, providing a theoretical basis for further testing of GM-CSF as a non-invasive treatment for COVID-19.

Mechanistic analysis and significance of sphingomyelinase-mediated decreases in transepithelial CFTR currents in nHBEs

Kirsten A. Cottrill et al.

Summary: Loss of function of CFTR due to acute bacterial SMase increases dihydroceramide abundance, leading to sphingolipid imbalance in CF. Sphingomyelins, ceramides, and dihydroceramides are significantly elevated in CF cells compared to non-CF cells.

TGF-β-induced α-SMA expression is mediated by C/EBPβ acetylation in human alveolar epithelial cells

Hui Ding et al.

Summary: The study confirmed that deacetylation of C/EBP beta in A549 cells successfully ameliorated TGF-beta 1-induced EMT, as shown by reduction in alpha-SMA expression and excessive collagen-I accumulation. This indicates that the EMT and fibrotic effect of TGF-beta 1 depend on acetylated C/EBP beta-mediated regulation of alpha-SMA gene activity.

Chronic Alcohol Exposure of Cells Using Controlled Alcohol-Releasing Capillaries

Wanil Kim et al.

Summary: The study developed a glass capillary system containing ethanol to mimic chronic alcohol exposure conditions, leading to increased cell toxicity and altered cellular physiologies. This system could be a valuable tool for studying the mechanism of chronic alcohol-mediated hepatic diseases at the cellular level.
