Microenvironmental modulation in tandem with human stem cell transplantation enhances functional recovery after chronic complete spinal cord injury

Shogo Hashimoto et al.

Summary: In this study, the therapeutic effect of environmental modulation on severe chronic spinal cord injury (SCI) was explored. The use of a clinically relevant hepatocyte growth factor (HGF)-releasing scaffold and human iPS cell-derived neural stem/progenitor cell (hNS/PCs) transplantation led to functional recovery. This study provides evidence for the feasibility of regenerative medicine strategies using clinically relevant materials in the treatment of severe chronic SCI.

Linquan Zhou et al.

Summary: This study developed a photocurable silk fibroin hydrogel to deliver basic fibroblast growth factor (bFGF) for spinal cord injury treatment. The hydrogel exhibited slow release of bFGF, promoting nerve axon regeneration, inhibiting glial cell proliferation, improving neuronal mitochondrial function, and suppressing local inflammation and oxidative stress. This novel method provides a new treatment strategy for spinal cord injury.

Restoration of spinal cord biophysical microenvironment for enhancing tissue repair by injury-responsive smart hydrogel

Caixia Fan et al.

Summary: Spinal cord injury is a severe damage to the central nervous system, often leading to the loss of spinal cord structure and function. Researchers have developed a new hydrogel material with mechanical and electrical properties similar to the spinal cord, which can be used for spinal cord regeneration. By introducing glutathione and MMP-responsive proteins into the hydrogel, biomolecules can be released in response to the microenvironment of spinal cord injury, promoting axon regeneration and angiogenesis, and improving motor function recovery.

Activity-dependent plasticity and spinal cord stimulation for motor recovery following spinal cord injury

Soshi Samejima et al.

Summary: Spinal cord injuries often result in permanent physical impairments despite being incomplete disruptions. However, remaining connections between the brain and spinal cord can induce neural plasticity to improve sensorimotor function, even years post-injury. This review provides an overview of evidence for motor recovery, plasticity, and interventions in spinal cord stimulation for motor control restoration. It discusses both open-loop and closed-loop stimulation approaches, as well as mechanisms of spinal cord neuromodulation for sensorimotor recovery, aiming to advance rehabilitation for spinal cord injuries.

Bi-Qin Lai et al.

Summary: For millennia, adult mammals have been unable to recover function lost after severe spinal cord injury due to a failure of brain-derived nerve fiber regeneration across the lesion. Various potential approaches rely on neuronal relays to reconnect the segregated neural networks of the spinal cord, including the use of stem cell-derived neuronal relays and functional electrical stimulation.

Self-assembling peptide hydrogels for the stabilization and sustained release of active Chondroitinase ABC in vitro and in spinal cord injuries

Andrea Raspa et al.

Summary: After spinal cord injuries, harmful events such as activated microglia/macrophages infiltration, astrocyte migration, and increased production of inhibitory CSPGs often occur. Chronic SCI results in a gliotic scar that acts as a barrier to axonal regrowth, but biomaterial scaffolds show promise in promoting nervous cell restoration. Research focusing on self-assembling peptide (SAP) hydrogels loaded with Chondroitinase ABC (ChABC) demonstrates potential for enhancing host neural regeneration and behavioral recovery in chronic SCI.
