Accurate and Early Metastases Diagnosis in Live Animals With Multimodal X-ray and Optical Imaging

Jiahao Chen et al.

Summary: The article introduces a research platform called iSMAART, which integrates CT and optical tomography for disease tracking and therapeutic evaluation in animal studies. The iSMAART system accurately captures 3D image information by using CT for anatomical and structural information, and optical tomography for tumor imaging. The system is validated in a prostate cancer metastasis model and achieves accurate tumor imaging and quantitative assessment. With the integration of CT and optical tomography, iSMAART has the potential to contribute to more complex tumor metastasis research.

Novel, highly sensitive, in vivo screening method detects estrogenic activity at low doses of bisphenol A

Keishi Ishida et al.

Summary: We developed a new short-term screening test using in vivo bioluminescence imaging to detect chemicals with estrogen-like effects. Our method is more sensitive and requires less time compared to traditional in vivo screening methods. Our results showed that our method could detect the estrogenic effects of BPA at doses below tolerable daily intakes, which traditional screening methods could not. Our study demonstrates that in vivo bioluminescence imaging is a highly effective tool for screening estrogenic chemicals and resolving the controversy about low-dose effects of endocrine-disrupting chemicals.

Crystal structure of semisynthetic obelin-v

Marina D. Larionova et al.

Summary: Coelenterazine-v (CTZ-v) is a suitable substrate for deep tissue imaging with Renilla luciferase, but Ca2+-regulated photoproteins activated by CTZ-v have low bioluminescence activity and are unsuitable for calcium imaging. The low bioluminescence activity of obelin-v is attributed to the low efficiency of generating an electronic excited state, possibly due to small changes in the substrate microenvironment at the active site.

Theoretical study on the radical scavenging activity and mechanism of four kinds of Gnetin molecule

Yaxuan Shang et al.

Summary: As an important subgroup of resveratrol oligomers, Gnetins have attracted much attention due to their antioxidant properties. Through thermodynamic and kinetic analysis using density functional theory, it was found that the antioxidant activity of Gnetins is related to their structure, with type A structure exhibiting higher radical scavenging activity.

Tuning the Intramolecular Chemiexcitation of Neutral Dioxetanones by Interaction with Ionic Species

Carla M. Magalhaes et al.

Summary: In this study, TD-DFT methodology was used to evaluate the control of intramolecular chemiexcitation of model dioxetanones by external stimuli. It was found that the addition of ionic species near neutral dioxetanones reduced the activation barrier of the thermolysis reaction, enabling dark catalysis. While the inclusion of these species negatively affected the chemiexcitation profile compared to anionic dioxetanones, their efficiency was at least comparable.

Chemistry in Fungal Bioluminescence: A Theoretical Study from Luciferin to Light Emission

Ming-Yu Wang et al.

Summary: Bioluminescent fungi can emit green light continuously for days and share a common mechanism among all lineages. Detailed processes remain unknown, but theoretical calculations have provided insights into the molecular and electronic state level of the bioluminescent process. The initiation of fungal bioluminescence involves the formation of a high-energy intermediate through oxygenation of luciferin.

Theoretically obtained insight into the effect of basic amino acids on Cypridina bioluminescence

Chun-Xia Liu et al.

Summary: Using theoretical Density Functional Theory (DFT) and Time Dependent Density Functional Theory (TDDFT) approaches, this study investigated the solvent effect on the fluorescent spectrum, the basicity of amino acids, and the covalent character of hydrogen bonds on Cypridina oxyluciferin. Results showed that the emission wavelengths were minimally affected by solvent polarity, and the effects of basic amino acids and environmental polarity on the structure and spectroscopic properties were slight. The covalent character of the hydrogen bond was found to modulate the fluorescent properties of Cypridina oxyluciferin.

H2O-Bridged Proton-Transfer Channel in Emitter Species Formation in Obelin Bioluminescence

Shu-Feng Chen et al.

Summary: The bioluminescence of marine organisms is regulated by CaRP, with the formation of the emitter involving a proton transfer (PT) process. Studies have shown that there is a concerted PT process between the anionic S-1-coelenteramide and the nearest histidine residue, facilitated by a water molecule. This fast PT process helps to align experimental and theoretical findings in emitter formation for CaRPs.
