Pilot study suggests DNA methylation of the glucocorticoid receptor gene (NR3C1) is associated with MDMA-assisted therapy treatment response for severe PTSD

Candace R. Lewis et al.

Summary: This study found that MDMA-assisted therapy may induce DNA methylation changes in HPA genes, leading to improvement in symptoms of post-traumatic stress disorder. These findings can be used to generate hypothesis driven analyses for future studies with larger cohorts.

Face your fears: attenuating remote fear memories by reconsolidation-updating

Bianca A. Silva et al.

Summary: This review examines the limited evidence on remote fear memory attenuation from animal and human research. It is found that although remote fear memories are harder to change compared to recent memories, they can still be attenuated through interventions targeted at the reconsolidation window during memory recall. The physiological mechanisms underlying remote reconsolidation-updating approaches are described, along with the potential enhancement through interventions promoting synaptic plasticity. By utilizing a relevant phase of memory, reconsolidation-updating has the potential to permanently alter remote fear memories.

Understanding the physical basis of memory: Molecular mechanisms of the engram

Clara Ortega-de San Luis et al.

Summary: Memory is the storage and use of learned information in the brain, which is crucial for animals to adapt and survive. The molecular and cellular mechanisms of memory formation are still not fully understood. The engram theory suggests that a group of active neurons form a trace during learning, undergo changes to store information, and are later reactivated during memory recall. Engram technology has allowed researchers to study memory formation at a molecular and cellular level, providing insights into different memory processes.

A randomized controlled trial of 3,4-methylenedioxymethamphetamine (MDMA) and fear extinction retention in healthy adults

Jessica L. Maples-Keller et al.

Summary: This study investigated the effects of MDMA on fear learning, extinction training, and retention in healthy humans. Although the hypothesized facilitation of extinction retention was not observed, the findings from this initial human trial provide compelling rationale to continue exploring the potential impact of MDMA on extinction retention.

MDMA treatment paired with a trauma-cue promotes adaptive stress responses in a translational model of PTSD in rats

Shira Arluk et al.

Summary: This study aimed to assess the bio-behavioral underpinnings of MDMA in a translational model of PTSD. The results showed that MDMA treatment paired with memory reactivation by a trauma-cue effectively attenuated stress behavioral responses and normalized the dendritic cytoarchitecture of DG and BLA neurons. These anxiolytic-like effects seem to involve the HPA axis and 5-HT systems.

Summary: Epigenetic research in PTSD examines the role of environmental stressors and fear in the disorder's development, providing insights into individual differences in prevalence and treatment response. The review discusses epigenetic markers associated with PTSD and treatment, as well as animal research on fear memory acquisition, update, and erasure. It also highlights the potential use of reconsolidation blockade as a treatment for PTSD and the challenges in studying epigenetics in humans.

Summary: This study aims to explore the potential of MDMA as a treatment for posttraumatic stress disorder (PTSD) and its mechanism of action. The results show that MDMA treatment can enhance fear extinction learning and recall, but this effect may be limited to certain forms of learned fear responses. Additionally, MDMA does not influence fear reactions to conditioned cues.

Summary: Despite the availability of numerous antiseizure medications, a significant number of patients do not respond to these medications. Refractory seizures are common in patients with epileptic encephalopathies, emphasizing the need for more effective and safer medications. Recently, the repurposing of the anti-obesity medication fenfluramine has shown promising results for seizures associated with specific syndromes. However, a comprehensive review of fenfluramine is needed to optimize its clinical use.

Psychedelics and Neuroplasticity: A Systematic Review Unraveling the Biological Underpinnings of Psychedelics

Cato M. H. de Vos et al.

Summary: Clinical studies suggest that psychedelics induce rapid changes in neuroplasticity mechanisms, leading to alterations in gene and protein expression, increased neurotrophic factor levels, and enhanced dendritic complexity. Repeated administration of psychedelics can directly stimulate neurogenesis and elevate BDNF mRNA levels, indicating potential long-lasting effects on brain plasticity. Future research could focus on deciphering specific cellular mechanisms activated by different psychedelics to better understand their therapeutic potential.
