Astrocyte Immune Functions and Glaucoma

Summary: Astrocytes, a type of glial cells in the nervous system, play crucial roles in regulating the physiological functions of the central nervous system. In diseases like glaucoma, astrocytes can undergo phenotypic changes and exhibit immune functions, which are associated with various pathological events such as tissue remodeling, neuronal death, and infiltration of immune cells. This review focuses on discussing the involvement of astrocyte-lineage cells' immune functions in the pathogenesis of glaucoma.

The genetics of glaucoma: Disease associations, personalised risk assessment and therapeutic opportunities-A review

Summary: Glaucoma refers to a group of diseases characterized by progressive loss of retinal ganglion cells and associated visual field loss. Both early-onset and adult-onset forms of the disease have a strong genetic component. In this article, we summarize the known genetic associations for different forms of glaucoma and discuss the potential functional roles of these genes in disease pathogenesis. We also explore efforts to apply genetic knowledge to clinical practice, including gene-based tests for disease diagnosis and risk stratification, as well as gene-based therapies.

Glaucoma and biomechanics

Summary: This review summarizes recent studies on the role of biomechanics in glaucoma. It is found that various tissues, including the trabecular meshwork, cornea, peripapillary sclera, optic nerve head/sheath, and iris, have biomechanical involvement in different forms of glaucoma. Multiple mechanosensory mechanisms and signaling pathways are identified in the trabecular meshwork and optic nerve head. Biomechanical information has also been incorporated into glaucoma prognoses. However, the understanding of the role of biomechanics in glaucoma is still incomplete.

Molecular regulation of neuroinflammation in glaucoma: Current knowledge and the ongoing search for new treatment targets

Summary: Neuroinflammation plays a significant role in the neurodegenerative processes of glaucoma, with glial cells and peripheral immune cells causing toxicity to retinal ganglion cells. The interaction between glial cells and immune cells is crucial in amplifying neuroinflammation and determining the neurodegenerative outcome. Inhibition of neurodegenerative inflammation may provide a broad treatment strategy for rescuing retinal ganglion cell integrity and function, while targeting both inflammatory toxicity and mitochondrial dysfunction.

Summary: The study showed that B6.EDA(+/+) mice exhibit increased ECM accumulation in the TM, elevated IOP, enhanced proinflammatory changes in the ONH, loss of RGCs, and ONH damage. These data suggest that B6.EDA(+/+) mice recapitulate many aspects of glaucomatous damage.

Cross-talk between GABAergic postsynapse and microglia regulate synapse loss after brain ischemia

Summary: This study investigates the role of microglia in synapse elimination after ischemia and identifies brain-derived neurotrophic factor (BDNF) as a key player in this process. The researchers found that microglia release BDNF to downregulate glutamatergic and GABAergic synapses in the peri-infarct area. They also discovered that microglia activation and gephyrin phosphorylation are involved in neuroplasticity after middle cerebral artery occlusion (MCAO). Deletion of the Bdnf gene in microglia or GphnS268A/S270A mutation protects against brain damage, neuroinflammation, and synapse downregulation after MCAO.

It is found that astrocytes play crucial roles in processes such as neuroinflammation and mitochondrial dysfunction, and therapeutic strategies targeting their beneficial or detrimental reactivity are proposed for future glaucoma treatment.

Summary: Glaucomatous optic neuropathy is the main cause of irreversible blindness worldwide. Lowering intraocular pressure is the only proven treatment, but it does not prevent further nerve degeneration. Cells in the optic nerve head, including lamina cribrosa cells, glial cells, and scleral fibroblasts, respond to glaucomatous conditions through extracellular matrix remodeling. Therefore, researching alternative therapies to alter this remodeling response is crucial for enhancing the survival of retinal ganglion cell axons.

Inflammation in Glaucoma: From the back to the front of the eye, and beyond

Summary: The pathophysiology of glaucoma is complex and multifactorial, involving elevated intraocular pressure and impaired retinal blood flow as primary factors. Oxidative stress, cell senescence, mitochondrial dysfunction, excitotoxicity, and neuroinflammation play crucial roles in glaucoma progression. Inflammatory processes in various ocular structures from the posterior to the anterior segment are present in glaucoma, supporting the hypothesis of dysregulation towards a pro-inflammatory phenotype. Possible anti-inflammatory therapeutic approaches for glaucoma are also discussed.

Rapid morphologic changes to microglial cells and upregulation of mixed microglial activation state markers induced by P2X7 receptor stimulation and increased intraocular pressure

Summary: The study investigated the morphological and molecular consequences of P2X7 receptor stimulation in microglial cells, both in vivo and in vitro, as well as the receptor's contribution in a model of increased intraocular pressure. Results showed that P2X7 receptor stimulation led to rapid activation of microglial cells, including process retraction and cell body enlargement, and upregulation of markers associated with M1 and M2 activation. The findings suggest that ATP release and P2X7 receptor stimulation play a role in the early microglial response to pressure changes.
