Mitochondrial energy metabolism correlates with an immunosuppressive tumor microenvironment and poor prognosis in esophageal squamous cell carcinoma

Single-Cell Transcriptomic Analysis of Primary and Metastatic Tumor Ecosystems in Esophageal Squamous Cell Carcinoma

Summary: In this study, the authors analyzed the transcriptomes of 85,263 single cells from four ESCC patients with lymph node metastasis. They identified various cells and genes that play a role in the metastatic microenvironment, including IFIT3(+) T and B cells, APOC1(+)APOE(+) macrophages, myofibroblasts, and immunoglobulin genes. They also discovered an epithelial-immune dual expression program that regulates immune processes in metastatic cells. Additionally, they found differential intercellular communications in the metastatic ESCC microenvironment, mainly involving the interaction of APOC1(+)APOE(+) macrophages with tumor and stromal cells. These findings provide insights into the molecular mechanisms underlying lymph-node metastasis and offer potential targets for inhibiting tumor growth and improving clinical outcomes.

Summary: Tumor cells promote the recruitment of immunosuppressive neutrophils, a subset of myeloid cells driving immune suppression, tumor proliferation, and treatment resistance. We have identified a subset of neutrophils that have upregulated expression of cellular senescence markers and persist in the tumor microenvironment. These senescent-like neutrophils express TREM2 and are more immunosuppressive and tumor-promoting than canonical immunosuppressive neutrophils.

LC3A-mediated autophagy regulates lung cancer cell plasticity

Summary: In this study, it was found that the key autophagy molecule MAP1LC3A/LC3A is negatively associated with histological grade and distant metastasis of lung cancer, partly due to its role in maintaining mitochondria and energy homeostasis. Basal autophagy is preferentially active in SOX2-positive lung cancer cells with high-proliferative and low-invasive properties. The findings provide insights into the crosstalk between basal autophagy and SOX2 proliferation signaling, regulating mitochondrial metabolism and determining cancer cell plasticity in lung tumor progression.

Diagnostic and prognostic values of pyroptosis-related genes for the hepatocellular carcinoma

Mutations in oncogenes, tumor suppressor genes, and metabolic enzymes alter various mitochondrial pathways in tumors, leading to metabolic reprogramming that sustains rapid cell proliferation and the utilization of reactive oxygen species by cancer cells. The knowledge acquired on mitochondrial cancer metabolism is now being translated into clinical practice, where detailed analysis of tumors is necessary to develop more precise treatments. Drugs targeting metabolic mitochondrial enzymes have shown potential in precision medicine, but efficient personalized drugs remain challenging to develop and may require combination with other strategies.
