Calycosin-7-glucoside promotes mitochondria-mediated apoptosis in hepatocellular carcinoma by targeting thioredoxin 1 to regulate oxidative stress

Xiaodong Wei et al.

Summary: In this study, the flavonoid calycosin-7-glucoside (CG) was investigated for its anti-hepatocellular carcinoma (HCC) activity by targeting thioredoxin1 (TRX1). Results demonstrated that CG dose-dependently decreased proliferation and induced apoptosis in HCC cells, while activating oxidative stress and inhibiting TRX1 expression. In vivo experiments also showed that CG regulated oxidative stress and TRX1 expression, and inhibited HCC growth. Molecular docking confirmed the binding effect between CG and TRX1. It was concluded that CG exerts its anti-HCC activity by targeting TRX1 to regulate oxidative stress and promote mitochondria-mediated apoptosis.

The therapeutic mechanism of Yuye decoction on type 2 diabetes mellitus based on network pharmacology and experimental verification

Feng Guo et al.

Summary: This study systematically explored the potential mechanism of Yuye decoction (YYD) against type 2 diabetes mellitus (T2DM) using UHPLC-MS/MS, network pharmacology, molecular docking, and experimental validation. The results suggest that YYD may prevent pancreatic islet dysfunction and reverse β-cell apoptosis through PI3K/AKT1 and Sirt1/FoxO1 signaling pathways. This study provides scientific evidence for the clinical application of YYD in the treatment of T2DM.

Contemporary medical, device, and surgical therapies for obesity in adults

Carolina M. Perdomo et al.

Summary: The goal of obesity management is to improve health, and sustained weight loss of more than 10% of overall bodyweight can achieve this. Lifestyle interventions, anti-obesity medications, and bariatric surgery are effective strategies to maintain weight loss and improve associated health outcomes. Recent therapeutic advances have led to the development of targeted treatments for monogenic obesities and a new generation of highly effective anti-obesity medications. Long-term studies are needed to assess the safety, efficacy, and cardiovascular outcomes of these treatments. Additionally, further research is required to determine optimal treatment strategies for individual patients and ensure access to effective treatments.

Gentiana lutea attenuates hepatotoxicity induced by ketoconazole in rats by fortifying the cellular antioxidant defense system

Alaaeldin Ahmed Hamza et al.

Summary: In this study, the hepatoprotective effects of Gentiana extract (GEN) against ketoconazole (KET)-induced hepatotoxicity were investigated in male Wistar rats. The results showed that GEN reduced liver damage induced by KET, as evidenced by decreased levels of aminotransferase and alpha-fetoprotein in serum and improved hepatic histopathological changes. Furthermore, GEN inhibited oxidative stress induced by KET by reducing lipid peroxidation, protein carbonylation, and oxidative stress in DNA. These findings suggest that GEN's hepatoprotective effects are attributed to its antioxidant properties.

Augmenter of liver regeneration: Mitochondrial function and steatohepatitis

Alok Kumar Verma et al.

Summary: Augmenter of liver regeneration (ALR) is a protein that is expressed more abundantly in the liver than other organs. It plays important roles in protein folding, cytochrome c reductase activity, and protein Fe/S maturation. ALR deficiency in hepatocytes leads to oxidative stress, impaired ATP synthesis, and increased apoptosis/necrosis. Individuals with one functional ALR allele may be susceptible to disorders involving compromised mitochondrial function.

Control of mitochondrial dynamics and apoptotic pathways by peroxisomes

Chenxing Jiang et al.

Summary: Peroxisomes are organelles that play a role in various metabolic pathways. Their biogenesis is controlled by a group of proteins, and mutations in these proteins can cause metabolic disorders. Recent research has shown that peroxisomes are involved in regulating cell apoptosis and fusion processes, and disruption of these processes may be related to the pathogenesis of specific diseases.

Obese visceral fat tissue inflammation: from protective to detrimental?

Hubert Kolb

Summary: Obesity-induced inflammation of adipose tissue is initially a protective response, but may contribute to tissue damage later on. The metabolic imbalance of adipocytes in obesity leads to activation of resident immune cells, enhancing tissue protective responses. However, chronic overnutrition results in insufficient protective actions, leading to adipocyte death and cellular senescence. This can cause influx and activation of immune cells, as well as intensified matrix remodeling and angiogenesis.

Metformin effectively restores the HPA axis function in diet-induced obese rats

Andrew C. Shin et al.

Summary: This study demonstrated that oral metformin can effectively reduce weight gain, fat mass, and caloric intake in obesity-prone rats. Metformin dose-dependently decreased HPA axis activity, normalized NE-HPA axis coupling, and reduced circulating free fatty acids levels in these rats. Effect of traditional Chinese medicine on gut microbiota in adults with type 2 diabetes: A systematic review and meta-analysis

Yujiao Zheng et al.

Summary: Recent studies have shown the significant role of gut microbiota in the development of type 2 diabetes mellitus (T2DM) and the potential therapeutic effect of traditional Chinese medicine (TCM) on T2DM. This systematic review included five randomized controlled trials (RCTs) on TCM interventions targeting gut microbiota in T2DM patients, showing statistically significant improvements in HbA1c, fasting blood glucose, postprandial blood glucose, insulin resistance, and beta-cell function. While TCM was found to modulate gut microbiota and improve glucose metabolism, further studies are needed to explore its effects on inflammation.

PRAP1 is a novel lipid-binding protein that promotes lipid absorption by facilitating MTTP-mediated lipid transport

Hubert Peng et al.

Summary: MTTP is essential for assembling and secreting triglyceride-rich lipoproteins, and a newly identified protein PRAP1 plays a key role in MTTP-mediated lipid transport. PRAP1 binds to triglycerides and facilitates lipid transfer alongside MTTP. Deficiency or mutation of PRAP1 leads to challenges in lipid absorption, but reduces body weight and fat mass on high-fat diets.
