Enabling Genetic Code Expansion and Peptide Macrocyclization in mRNA Display via a Promiscuous Orthogonal Aminoacyl-tRNA Synthetase

Summary: mRNA display revolutionizes peptide drug discovery by quickly identifying potent peptide binders for therapeutic protein targets. This study demonstrates the use of promiscuous orthogonal aminoacyl-tRNA synthetases (ORSs) in in vitro translation (IVT) and mRNA display to expand side chain diversity and enable heterocycle forming macrocyclization. The resulting macrocyclic binder exhibits good selectivity for USP15 and close homologues, highlighting the potential of ORSs in peptide ligand discovery.

Chemical Synthesis and Cell-Free Expression of Thiazoline Ring-Bridged Cyclic Peptides and Their Properties on Biomembrane Permeability

Inhibition of H1 and H5 Influenza A Virus Entry by Diverse Macrocyclic Peptides Targeting the Hemagglutinin Stem Region

Summary: This study reports a set of macrocyclic peptides that inhibit influenza A virus infection by binding to hemagglutinin. These peptides have broad-spectrum activity against different variants of influenza A virus and do not show cytotoxicity.
