Bioinspired Jellyfish-like Carbon/Manganese nanomotors with H2O2 and NIR light Dual-propulsion for enhanced tumor penetration and chemodynamic therapy

Summary: Chemodynamic therapy (CDT), which catalyzes the production of highly toxic reactive oxygen species (ROS), has been widely used in tumor therapy. However, the effectiveness of CDT is often limited by the poor tumor penetration of current nanomaterials. In this study, bioinspired jellyfish-like carbon/manganese nanomotors (JCMNs) were designed to achieve dual-propulsion for enhanced CDT therapy. The JCMNs utilized hydrogen peroxide (H2O2) and NIR light to propel themselves and improve tumor penetration and ROS generation, leading to improved CDT efficiency.

Co-delivery of doxorubicin/sorafenib by DNA-decorated green ZIF-67-based nanocarriers for chemotherapy and hepatocellular carcinoma treatment

Summary: In this study, natural biomaterials and DNA were successfully decorated on ZIF-67 to develop a promising co-delivery platform for the drug DOX-SOR. The modified MOFs showed stable structure and efficient drug loading and release. Cell experiments demonstrated that the decorated nanoparticles not only improved cell viability but also showed anticancer properties.

Pulmonary delivery of curcumin and quercetin nanoparticles for lung cancer-Part 2: Toxicity and endocytosis

Summary: The delivery of polyphenols, such as curcumin and quercetin, via inhalation can effectively treat lung cancer by increasing drug concentration in tumor cells. However, their applications have been limited by poor solubility and stability. In this study, inhaled curcumin nanoparticles and quercetin nanoparticles were shown to have enhanced anti-cancer activity in A549 cells compared to their individual formulations. Higher ratios of quercetin in the combination formulations induced more apoptosis, reduced ATP levels, generated more ROS, and caused MMP loss in A549 cells. The combination formulations also increased the expression of pro-apoptotic protein Bax. The toxicity of the formulations followed the trend: Cur-NPs: Que-NPs (1:4) > Cur-NPs: Que-NPs (2:3) > Cur-NPs: Que-NPs (3:2) > Cur-NPs: Que-NPs (4:1) > Cur-NPs only > Que-NPs only. Furthermore, both Cur-NPs and Que-NPs were internalized by cells through clathrin-mediated endocytosis. Overall, the combination of Cur-NPs and Que-NPs delivered via inhalation may serve as an alternative treatment for lung cancer.

Self-Propelled Janus Nanocatalytic Robots Guided by Magnetic Resonance Imaging for Enhanced Tumor Penetration and Therapy

Summary: In this study, the integration of micro/nanorobots and nanocatalytic medicine was used to achieve self-propulsion-induced deeper tumor penetration and catalytic reaction-initiated tumor therapy in vivo. The self-propelled Janus nanocatalytic robots (JNCRs) were guided by magnetic resonance imaging (MRI) for active movement and their migration in the tumor tissue could be tracked by non-invasive MRI in real time. These JNCRs showed great potential for tumor photothermal and chemodynamic therapy through near-infrared light irradiation and iron-mediated Fenton reaction.

Magnetotactic Bacteria-Based Drug-Loaded Micromotors for Highly Efficient Magnetic and Biological Double-Targeted Tumor Therapy

Summary: Bacteria-mediated cancer therapy has gained attention, but using magnetotactic bacteria for therapy has not been reported. In this study, a photosensitizer was incorporated into a bacterial strain M. magneticum AMB-1, creating AMB-1/Ce6 micromotors with high drug payload and bacterial activities. It was found that AMB-1 not only acts as a drug carrier but also inhibits tumor growth. Under laser irradiation, micromotors efficiently killed cancer cells by generating reactive oxygen species. They could target hypoxic and normoxic regions through active swimming and magnetic field guidance, showing high tumor-homing ability. The micromotors significantly inhibited mouse tumor growth through biotherapy and phototoxicity. This strategy may be a promising platform for treating solid tumors.

Micromotor Based Mini-Tablet for Oral Delivery of Insulin

Summary: A mini-tablet system based on self-propelled micromotors was successfully developed for oral colon administration of insulin. The system achieved controlled release of insulin through the degradation of esterified starch coating. The released micromotors, with autonomous movement, significantly enhanced colonic uptake and absorption of insulin, leading to long-term blood glucose control. This system not only expands the biomedical applications of self-propelled micromotors but also provides an appealing strategy for oral administration of biomacromolecular drugs.

ROS-responsive magnesium-containing microspheres for antioxidative treatment of intervertebral disc degeneration

Summary: Antioxidant therapy is a promising strategy for intervertebral disc degeneration (IVDD) treatment due to the crucial role of reactive oxygen species (ROS) in IVDD pathogenesis. In this study, ROS-responsive magnesium-containing microspheres (Mg@PLPE MSs) were developed for the controlled release of hydrogen gas to alleviate IVDD. The Mg@PLPE MSs effectively scavenged overproduced ROS and created a suitable microenvironment for inhibition of extracellular matrix (ECM) degradation. In an IVDD rat model, the Mg@PLPE MSs demonstrated significant antioxidative and anti-inflammatory effects, reducing ECM degradation and disc cell apoptosis. They also showed biocompatibility and low toxicity, holding promise for clinical IVDD treatment.

Co-Delivery of Loxoprofen and Tofacitinib by Photothermal Microneedles for Rheumatoid Arthritis Treatment

Summary: By fabricating a dissolving microneedle system with photothermal polydopamine, the transdermal co-delivery of the anti-inflammatory drug loxoprofen and Janus kinase inhibitor tofacitinib directly to the articular cavity was achieved. This approach effectively reduced symptoms of rheumatoid arthritis, such as joint swelling, muscle atrophy, and cartilage destruction, and downregulated proinflammatory cytokine expression levels. The photothermal microneedle system offers high compliance and good therapeutic efficacy.

Gastric acid powered micromotors for combined-drug delivery to eradiate helicobacter pylori

Summary: We designed an active drug combination therapy and responsive drug release for Helicobacter pylori using self-propelled micromotors. These micromotors can penetrate the mucus layer, deliver amoxicillin and clarithromycin, and release them in a pH-responsive manner. The micromotors showed enhanced retention and distribution capability, and significantly improved antimicrobial efficiency compared to single-drug loaded micromotors. The optimized dosage ratio of amoxicillin and clarithromycin on the micromotors was determined to be 2:1.

Dual-propelled PDA@MnO2 nanomotors with NIR light and H2O2 for effective removal of heavy metal and organic dye

Summary: This study optimized niosomes for carrying and delivering anticancer agents and genes, showing that co-delivery of curcumin and miR-34a using the new cationic niosomal delivery system can be recognized as a prominent strategy for more effective cancer treatments. Cancer cells treated with co-delivery showed higher cytotoxicity, uptake, and anticancer activity, while showing less toxicity to normal human cells. In addition, the co-delivery of curcumin and miR-34a in the 4T1 xenografted Balb/C mouse tumor model resulted in a higher tumor inhibition rate compared to other groups.

Gastrointestinal tract drug delivery using algae motors embedded in a degradable capsule

Magnesium-Based Micromotors as Hydrogen Generators for Precise Rheumatoid Arthritis Therapy
