Quantification of gallium cryo-FIB milling damage in biological lamellae

Bronwyn A. Lucasa et al.

Summary: Cryogenic electron microscopy (cryo-EM) can reveal molecular details of biological processes with atomic resolution, but thinning of cells for cryo-EM imaging is limited. Focused-ion-beam (FIB) milling has enabled visualization of cellular structures by thinning frozen cells to <500 nm lamellae. However, the damage caused by FIB milling to cell sections has not been determined.
