Metalloradical approach for concurrent control in intermolecular radical allylic C-H amination

Summary: Co(II)-based metalloradical catalysis enables the chemoselective intermolecular amination of allylic C-H bonds and achieves remarkable convergence of regioselectivity, diastereoselectivity, and enantioselectivity. The use of modularly designed D-2-symmetric chiral amidoporphyrins as supporting ligands creates a tunable pocket-like environment. This reaction offers a direct route to synthesize valuable chiral alpha-tertiary amines from an isomeric mixture of alkenes.

Pd-Catalyzed Asymmetric Allylic C-H Functionalization

Summary: Pd-catalyzed asymmetric allylic C-H functionalization is a powerful tool to access chiral and densely functionalized molecules from easily accessible alkenes, resulting in increased step- or atom-economy by minimizing functional group manipulations. However, due to inadequate stereoselection strategies, this field is still in its early stage. This essay describes the journey to identify asymmetric catalytic systems, the mechanism of allylic C-H activation, control of stereo- and regioselectivity, and applications in asymmetric synthesis.

Palladium-Catalyzed Branch- and Z-Selective Allylic C-H Amination with Aromatic Amines

Summary: Allylamines are important for synthesizing bioactive compounds and can be obtained through the palladium-catalyzed allylic C-H amination of 1,4-dienes. This reaction favors the formation of thermodynamically unstable Z-allylamines with aromatic amines. Aliphatic cyclic amines can also participate, but linear dienyl allylic amines are the main products.

Stereodefined Skipped Dienes through Iridium-Catalyzed Formal Addition of Tertiary Allylic C-H Bonds to Alkynes

Summary: We report an iridium-catalyzed method for the facile preparation of skipped dienes with a quaternary carbon center at the C-3 position. The method involves the formal addition of a tertiary sp(3) C-H bond to an alkyne, cleaving the tertiary allylic C-H bond selectively and forming a new C-C bond. Additionally, enantioselective construction of acyclic quaternary carbon stereocenters is demonstrated.

Asymmetric intermolecular allylic C-H amination of alkenes with aliphatic amines

Summary: Aliphatic allylic amines are complex and biorelevant molecules that are present in a wide range of compounds. The direct allylic C-H amination of alkenes is the simplest method to obtain these motifs. However, using widely available internal alkenes with aliphatic amines remains a synthetic challenge. We report a general solution to these problems using a blue light-induced open-shell approach and a mild oxidant.

Palladium-Catalyzed Enantioselective C(sp3)-H/C(sp3)-H Umpolung Coupling of N-Allylimine and α-Aryl Ketones

Summary: Asymmetric functionalization of the C(sp(3))-H bond using chiral phosphoramidite-palladium catalysis was successfully achieved in this study, leading to the synthesis of enantioenriched beta-amino-gamma,delta-unsaturated carbonyl derivatives and providing a key intermediate for the asymmetric synthesis of Focalin. The research also revealed an umpolung reactivity of N-allylimines through a concerted proton and two-electron transfer process.

The catalytic system exhibited high branch selectivity and a wide substrate scope, leading to the synthesis of a useful intermediate for (R)-alpha-lipoic acid.
