Improving the hydrolysis and acyltransferase activity of bifunctional feruloyl esterases DLFae4 by multiple rational predictions and directed evolution

Testing and improving the performance of protein thermostability predictors for the engineering of cellulases

Summary: Thermostability of cellulases can be improved by amino acid substitutions and protein engineering with thermostability predictors. We analyzed the performance of 18 predictors for cellulase engineering and found that DynaMut, SDM, RosettaDesign, and PremPS showed the highest accuracy, F-measure, and MCC values. The combination of predictors improved the performance, with a 14% increase in F-measure and 28% increase in MCC. Accuracy and sensitivity also increased by 9% and 20%, respectively, compared to the best single predictors. These findings can facilitate research in engineering thermostable cellulases and developing thermostability predictors.

Enhancing the Hydrolysis and Acyl Transfer Activity of Carboxylesterase DLFae4 by a Combinational Mutagenesis and In-Silico Method

Summary: In this study, a feruloyl esterase DLFae4 was modified through error-prone PCR and site-directed saturation mutation to enhance its catalytic efficiency and acyltransferase activity. Five mutants with enhanced catalytic activity towards methyl ferulate were characterized. Among them, the double variant DLFae4-m5 exhibited the highest hydrolytic activity, with a decrease in Km value and an increase in Kcat/Km value. Molecular docking revealed that DLFae4-m5 formed a complex hydrogen bond network, resulting in increased catalytic activity. Furthermore, the mutation of residue Ala-341 to glycine (A341G) in DLFae4 enhanced its acyl transfer activity and efficiency, representing the highest acyltransferase activity reported so far for family VIII carboxylesterases. These findings demonstrate the effectiveness of error-prone PCR and site-directed saturation mutation in improving enzyme activity and highlight the practical application of this feruloyl esterase.

Discovery and Design of Family VIII Carboxylesterases as Highly Efficient Acyltransferases

Summary: EstCE1, identified as an efficient acyltransferase catalyzing amidation and carbamoylation in water, provides a solution to the current challenges faced by promiscuous enzymes in application. The structure-function analysis of EstCE1 reveals a three-amino acid motif essential for its promiscuous acyltransferase activity. Introducing this motif into an esterase without acetyltransferase activity can transform it into an acyltransferase.
