A simple and green solid phase extraction method for oligonucleotides using adsorbent with amino and carboxylic moieties

Summary: This study aimed to extract unmodified and phosphor-othioate oligonucleotides using a material with amino and carboxylic moieties. The charge of the surface groups changed with pH, which allowed for the binding and elution of the oligonucleotides through electrostatic interactions. The method is simple, fast, reproducible, and environmentally friendly.

Effect of ion-pairing reagent hydrophobicity on liquid chromatography and mass spectrometry analysis of oligonucleotides

Summary: We systematically studied the performance of thirteen alkylamines as ion-pairing reagents for ion-pair reversed-phase liquid chromatography (IP RP LC) and proposed a method to classify the hydrophobicity of alkylamines. The results showed that alkylamines with higher hydrophobicity allowed for effective separation of oligonucleotides at lower buffer concentrations, and replacing acetic acid with hexafluoroisopropanol improved the mass spectrometry signal.

Development of the Method for Nusinersen and Its Metabolites Identification in the Serum Samples of Children Treated with Spinraza for Spinal Muscular Atrophy

Summary: The use of oligonucleotides as drugs for genetic diseases is increasing rapidly, and this study aimed to improve the analysis method for serum samples of patients treated with nusinersen. The results showed that a combination of liquid-liquid extraction and solid-phase extraction, along with optimizing the amine type in the mobile phase and the stationary phase, successfully identified multiple metabolites of nusinersen.

A perspective on oligonucleotide therapy: Approaches to patient customization

Summary: It is estimated that only 2% of disease-modifying proteins have a druggable site that can be targeted by approved clinical candidates. Therefore, there is a need to develop therapeutics that can mitigate diseases caused by both druggable and non-druggable proteins or enzymes. Oligonucleotide therapeutics (OT) have shown potential in this regard, as they have improved selectivity and fewer off-target effects compared to small molecules. This review provides an overview of the past and present of OTs, as well as the current efforts to develop future therapeutics. It also discusses the challenges and advancements in OT drug delivery and the emerging approach of personalized treatment for rare and fatal diseases.

Selectivity limits of and opportunities for ion pair chromatographic separation of oligonucleotides

Summary: In this study, it was investigated how oligonucleotide retention and selectivity in ion pair chromatography are affected by electrostatic and non-electrostatic interactions. It was found that selectivity increases with increasing surface potential and analyte charge difference for oligonucleotides of different charges. Additionally, selectivity for analytes of the same charge can be increased by decreasing co-solvent volume fraction or temperature.
