Polyfunctional donor-reactive T cells are associated with acute T-cell-mediated rejection of the kidney transplant

Nicolle H. R. Litjens et al.

Summary: Despite improvements in immunosuppressive regimens, acute TCMR remains a clinical problem after kidney transplantation. Polyfunctional donor-reactive T cells, specifically CD137++ T cells, are associated with the occurrence of a biopsy-proven aTCMR within the first year after transplantation. These cells are predominantly of the EM/EMRA-phenotype and show decreased polyfunctionality at the time of aTCMR.

Reinvestigation of Classic T Cell Subsets and Identification of Novel Cell Subpopulations by Single-Cell RNA Sequencing

Xuefei Wang et al.

Summary: Classic T cell subsets are defined by cell surface markers, while single-cell RNA sequencing clusters cells based on gene expression profiles. Multiple T cell subsets can be assigned to one cluster with similar expression patterns. Higher levels of ISAGs may contribute to quick immune responses.

Analysis of T-Cell Receptor Repertoire in Transplantation: Fingerprint of T Cell-mediated Alloresponse

Guangyao Tian et al.

Summary: This article reviews the recent advances in TCR sequencing techniques and computational tools, as well as the recent discovery in overall TCR profile and antigen-specific T cells tracking in transplantation. The challenges and potential of using TCR sequencing-based assays to profile alloreactive TCR repertoire as the fingerprint for immune monitoring and prediction of rejection and tolerance are further discussed.

Single cell multiomic analysis of T cell exhaustion in vitro

Mirko Corselli et al.

Summary: The study delved into the heterogeneity and plasticity of T cells under chronic stimulation, revealing varying proportions of cells expressing activation or exhaustion markers at different time points. Utilizing single-cell multiomic analysis, expression of 38 proteins and 399 genes in expanded human T cells was measured, leading to the identification of distinct activation states.

Progressive Loss of Donor-Reactive CD4+Effector Memory T Cells due to Apoptosis Underlies Donor-Specific Hyporesponsiveness in Stable Renal Transplant Recipients

Amy C. J. van der List et al.

Summary: This study investigates the characteristics of DSH, a lowered response of alloreactive T cells after kidney transplantation. The results show a decline in donor-reactive CD4+ T cells within the first 3-5 years after transplantation, mainly affecting effector memory T cells capable of producing proinflammatory cytokines. This decline is strongly correlated with reduced proliferation of CD4+ T cells to donor Ag.

Prospective Tracking of Donor-Reactive T-Cell Clones in the Circulation and Rejecting Human Kidney Allografts

Constantin Aschauer et al.

Summary: Sequencing of T-cell receptor repertoires in kidney transplant recipients revealed an enrichment of donor-reactive T-cells in the allograft during TCMR episodes, with dominant tissue clones also found in the blood. This study suggests an unchoreographed process of diverse T-cell clones directed against numerous non-self antigens in the transplanted organ.

Expression of Senescence Marker TIGIT Identifies Polyfunctional Donor-Reactive CD4+T Cells Preferentially Lost After Kidney Transplantation

Amy C. J. van der List et al.

Summary: The study aimed to investigate the decrease in responsiveness of CD4+ T cells to donor antigen post kidney transplantation, finding a significant decrease in the number of TIGIT-positive donor antigen-reactive T cells in elderly stable kidney transplant recipients.
