Trihydroxy piperlongumine protects aluminium induced neurotoxicity in zebrafish: Behavioral and biochemical approach

B. Haridevamuthu et al.

Summary: The present study investigates the antioxidant role of trihydroxy piperlongumine (THPL) against aluminum (Al)-induced neurotoxicity in zebrafish. THPL reduces oxidative damage in the brain by quenching Al-induced free radicals and lipid peroxidation. THPL also improves behavior deficits and anxiety-like phenotype in zebrafish, suggesting its potential as a psychopharmacological drug.

Hepatoprotective effect of dihydroxy piperlongumine in high cholesterol-induced non-alcoholic fatty liver disease zebrafish via antioxidant activity

B. Haridevamuthu et al.

Summary: Non-alcoholic fatty liver disease (NAFLD) and non-alcoholic steatohepatitis (NASH) are common liver diseases caused by a western diet high in fats, which induce inflammation and impair hepatocyte function. Currently, there is a lack of pharmacological drugs for treating NAFLD/NASH. However, long pepper contains a bioactive compound called Piperlongumine (PL) that has potential antioxidant properties. In this study, demethylated dihydroxy piperlongumine (DHPL) was shown to have high antioxidant activity and was effective in reducing lipid accumulation and gene expression associated with NAFLD in a zebrafish model. DHPL may have therapeutic potential for halting the progression of NAFLD.

Stage-Related Neurotoxicity of BPA in the Development of Zebrafish Embryos

Jianjun Liu et al.

Summary: BPA is a widely used chemical in the production of epoxy resins and polycarbonate plastics. This study found that BPA exposure impairs neural development and locomotor activity of zebrafish embryos. The key gene cypin is downregulated by BPA, and injecting cypin gene successfully restores locomotor activity in zebrafish.

Site-selective superassembly of biomimetic nanorobots enabling deep penetration into tumor with stiff stroma

Miao Yan et al.

Summary: The authors developed biomimetic head/hollow tail nanorobots that can efficiently remodel the dense tumor stroma in TNBC, enabling deep tumor penetration and improving cellular uptake efficiency. The nanorobots promote interfacial cellular uptake efficacy, transvascular extravasation, and intratumoral penetration through their self-movement ability and spiky head. These nanorobots effectively suppress tumor growth in a bone metastasis mouse model of TNBC and demonstrate potent antitumor efficacy in three different subcutaneous tumor models.

Effects of the food colorant carmoisine on zebrafish embryos at a wide range of concentrations

Tuba Kiziltan et al.

Summary: Since the middle of the twentieth century, the use of dyes has become more common in various industries. In this study, the effects of carmoisine on the embryonic development of zebrafish were investigated, while examining recommended and overexposure doses. The results showed that carmoisine caused serious malformations, reduction in height and eye diameter, increase in oxidative stress indicators and apoptosis, lipid accumulation, decreased locomotor activity and blood flow rate.

Hydroxyl containing benzo[b]thiophene analogs mitigates the acrylamide induced oxidative stress in the zebrafish larvae by stabilizing the glutathione redox cycle

B. Haridevamuthu et al.

Summary: Compounds BP and EP exhibited strong in vitro free radical scavenging ability. The maximum tolerated concentration of the compounds in zebrafish larvae was 80 μM. The antioxidant system in zebrafish larvae was dysregulated due to acrylamide exposure, and improvement was observed when acrylamide-exposed larvae were treated with compounds BP and EP. These compounds enhanced SOD and CAT activity, reduced ROS and lipid peroxidation levels, and decreased cell death in zebrafish larvae. Furthermore, BP and EP stabilized glutathione-related gene expressions, thus improving the glutathione redox cycle.

Amelioration of acrylamide induced neurotoxicity by benzo[b]thiophene analogs via glutathione redox dynamics in zebrafish larvae

B. Haridevamuthu et al.

Summary: This study demonstrates the therapeutic potential of compounds BP and EP in oxidative stress-associated neurological disorders. The experiment shows that these compounds can restore glutathione redox dynamics, reduce oxidative stress and neuroinflammation, and partially restore the normal function of motor neurons.

Withaferin A targets the membrane of Pseudomonas aeruginosa and mitigates the inflammation in zebrafish larvae; an in vitro and in vivo approach

Raghul Murugan et al.

Summary: Infections caused by multidrug-resistant Pseudomonas aeruginosa are common in patients with cystic fibrosis. Withaferin A extracted from ashwagandha has shown strong antibacterial activity against P. aeruginosa by damaging the cell membrane. Treatment with Withaferin A reduced oxidative stress and inflammatory response in a zebrafish larvae model infected with P. aeruginosa.

Neuroprotective effect of Biochanin a against Bisphenol A-induced prenatal neurotoxicity in zebrafish by modulating oxidative stress and locomotory defects

B. Haridevamuthu et al.

Summary: This study investigated the neuroprotective effect of Biochanin A (BCA) against Bisphenol A (BPA)-induced neuroinflammation. The findings suggested that co-exposure to BCA prevented the depletion of antioxidant defense enzymes by BPA and reduced the production of intracellular reactive oxygen species (ROS), thereby safeguarding neuronal development. BCA also improved motor neuron function, reduced proinflammatory response expression, and prevented neuroinflammation.

Tail Coiling Assay in Zebrafish (Danio rerio) Embryos: Stage of Development, Promising Positive Control Candidates, and Selection of an Appropriate Organic Solvent for Screening of Developmental Neurotoxicity (DNT)

Andreia A. S. de Oliveira et al.

Summary: The tail coiling assay in zebrafish embryos is proposed as an alternative method for screening developmental neurotoxicity induced by chemicals. Embryos from 22 to 29 hpf showed consistent coiling activity, with caffeine and MS-222 affecting activity levels differently. Using 1% DMSO as a solvent for the assay was considered relatively safe.
