Modern Approaches in the Discovery and Development of Plant-Based Natural Products and Their Analogues as Potential Therapeutic Agents

Asim Najmi et al.

Summary: Natural products are an important source of new lead compounds in drug discovery research, especially those derived from plants. Despite pharmaceutical companies' lack of attention to natural product drug discovery, recent technological advancements have revived scientific interest. This review provides a comprehensive overview of the various approaches used in selecting, authenticating, extracting/isolating, screening, and developing natural products, with a focus on bioactivity-guided fractionation. The integration of interdisciplinary approaches, technological advances, and new methods holds great potential for the future development of therapeutic drugs.

Extraction, preparative monomer separation and antibacterial activity of total polyphenols from Perilla frutescens

Yana Zhao et al.

Summary: Polyphenols, particularly RosA and CafA, were extracted from Perilla frutescens using ultrasonic-assisted ethanol extraction and ultrasound-assisted cellulase hydrolysis methods. The total phenolic content was significantly increased by cellulase hydrolysis, and further purification led to higher phenolic content. HPLC analysis revealed the composition of polyphenols, and a system for isolating high-purity RosA and CafA was established. Antibacterial activities of the polyphenols demonstrated effective inhibition of various bacteria.

Antibacterial activity and mechanism of three isomeric terpineols ofCinnamomum longepaniculatumleaf oil

Jinfeng Huang et al.

Summary: alpha-Terpineol, terpinen-4-ol, and delta-terpineol, isomers of terpineol, exhibit good inhibitory effects against several gram-negative bacteria, especially Shigella flexneri. They can increase membrane permeability and disrupt cell wall, leading to cell death.

Antioxidant, antimicrobial and anticancer activities ofCastanea sativa(Fagaceae) extract: new therapeutic perspectives

Carlo Genovese et al.

Summary: The study focused on investigating the antioxidant, antimicrobial, and anticancer properties of Castanea sativa ethanolic pellicle extract, which showed the highest total polyphenol and flavonoid content and exhibited the highest antioxidant activity. Only the ethanolic extract inhibited bacterial growth, while the aqueous extract showed no activity. The results suggest that the ethanolic pellicle extract of C. sativa may serve as a natural source of antioxidant, antimicrobial, and anticancer agents with potential health benefits.

Antimicrobial and anti-biofilm activity of thymoquinone against Shigella flexneri

Qiuxia Fan et al.

Summary: The study showed that thymoquinone has antimicrobial and anti-biofilm activities against Shigella flexneri by disrupting cell morphology, inhibiting protein synthesis, and downregulating genes involved in biofilm formation. Thymoquinone has the potential to be used as a natural agent to combat contamination and infection caused by Shigella flexneri.
