Summary: The production of chemicals and fuels from lignocellulosic biomass has great industrial potential, but the microorganisms used must be able to efficiently utilize all sugars in lignocellulosic hydrolysates. Although Yarrowia lipolytica is a good candidate for producing valuable products from biomass, it struggles to metabolize xylose efficiently. The development of metabolic engineering has enabled the bioconversion of xylose into various value-added products using Y. lipolytica as a platform.

A synthetic promoter system for well-controlled protein expression with different carbon sources in Saccharomyces cerevisiae

Summary: In this study, a library of synthetic GAL promoters was constructed, demonstrating a synergistic relationship between upstream activating sequences (UASs) and core promoters in determining promoter performance under different carbon sources. By manipulating UAS sequence, number, and substitution, the strength of synthetic GAL promoters could be fine-tuned. Core promoter replacement resulted in synthetic promoters with a twofold improvement in strength compared to the classic GAL1 promoter under multiple carbon sources in a strain with GAL1 and GAL80 engineering, expanding the dynamic range and carbon source tolerance of the GAL expression system.

Research investments have facilitated the simultaneous fermentation of xylose and glucose, with attention now turning towards improving xylose-utilizing efficiency and achieving robust co-fermentation under industrial conditions. Harnessing the advantageous traits of yeast xylose metabolism is expected to lead to the production of diverse fuels and chemicals.
