Affinity bioelectroanalysis in cellular-level biomarker driven modern precision cancer diagnosis

Summary: The role of liquid biopsy in precision, equitable, and sustainable diagnosis of prevalent and highly heterogeneous diseases using biomarkers derived from whole cells, secretomes, and exosomes is undeniable. Electrochemical biosensing and biosensors, particularly those involving affinity reactions, have emerged as competitive and cost-effective analytical alternatives for identifying these challenging targets. This review provides a concise and up-to-date overview of the fundamentals, versatility, and opportunities offered by state-of-the-art electrochemical biosensing (2018-2022) for the determination of cellular-derived biomarkers. It also offers critical insights into the short-term challenges and future research directions in this field.

First bioelectronic immunoplatform for quantitative secretomic analysis of total and metastasis-driven glycosylated haptoglobin

Summary: This work presents the first bioelectronic immunoplatforms for determining glycosylated haptoglobin (Hp) and both total and glycosylated Hp simultaneously. The immunoplatforms use non-competitive bioassays and magnetic microcarriers labeled with horseradish peroxidase (HRP) for detection. The method allows accurate determination of total and glycosylated Hp in colorectal cancer (CRC) cells, which is not possible with commercial ELISA kits and Western blot methodology due to lack of sensitivity.

Endostatin and Cancer Therapy: A Novel Potential Alternative to Anti-VEGF Monoclonal Antibodies

Summary: Angiogenesis is a process involving the formation of new blood vessels from preexisting ones, which plays important roles in physiological processes and tumor growth. Vascular endothelial growth factor (VEGF) is the main mediator of angiogenesis and is overexpressed in certain cancers, leading to the development of anti-VEGF monoclonal antibodies as cancer therapies. However, these antibodies have shown adverse events, while endostatin, an endogenous protein that inhibits VEGF expression and angiogenesis, has a better safety profile and promising results on small scale clinical studies. Therefore, endostatin is proposed as a replacement for anti-VEGF monoclonal antibodies.

A Cross-Sectional and Longitudinal Analysis of Pre-Diagnostic Blood Plasma Biomarkers for Early Detection of Pancreatic Cancer

Summary: This study proposes an effective method for early diagnosis of PDAC. By comparing different biomarkers and multivariable models, it was found that CA19-9 performed best within six months of diagnosis with high accuracy. Longitudinal analysis revealed a gradual increase in CA19-9 up to two years before diagnosis, providing an opportunity for early detection of PDAC.
