Improving cold-adaptability of mesophilic cellulase complex with a novel mushroom cellobiohydrolase for efficient low-temperature ensiling

Summary: A new cold-active cellobiohydrolase (VvCBHI-I) derived from Volvariella volvacea showed excellent activity at 10-30 degrees C, which enables efficient cellulose degradation and saccharification at low temperatures. The expression of VvCBHI-I in a transformant significantly improved the activity of cellulase at 10 degrees C, leading to stable silage fermentation with rice straw and reduction of cellulose and hemicellulose contents. These findings highlight the potential of mushroom-derived cold-active cellobiohydrolases in enhancing ensiling efficiency in cold climate regions.

Tetramerization of GH2 β-Glucuronidases is Essential for Catalyzing the Hydrolysis of the Large Substrate Glycyrrhizin

Summary: In this study, the researchers used structural analysis to identify three key residues that are crucial for the tetramer formation of a GH2 beta-glucuronidase enzyme from Aspergillus oryzae. Mutations at these sites disrupted the tetramer structure and resulted in loss of enzymatic activity. By refactoring a dimer into a tetramer using disulfide bonding, the researchers were able to partially restore the enzyme activity. Further experiments showed a strong correlation between the activity of the mutants and their tetramer ratios. Molecular simulations revealed the importance of the tetramer interface in maintaining the active-site structure. Finally, the researchers applied the identified tetramer-forming residues to other GH2 GUS enzymes and demonstrated the significance of homotetramerization for the enzymatic activity. This study lays the foundation for engineering GH2 GUS enzymes at the quaternary structure level to regulate their function.
