In vivo C6 glioma models: an update and a guide toward a more effective preclinical evaluation of potential anti-glioblastoma drugs

Mathematical modelling of the dynamics of image-informed tumor habitats in a murine model of glioma

Summary: In this study, the heterogeneity of tumors was captured using quantitative MRI data and described through imaging-based subregions or habitats in a murine model of glioma. The growth of these habitats was modeled and predicted using coupled ordinary differential equations (ODEs), both with and without radiotherapy. Three significantly different tumor habitats were identified based on imaging data characteristics: high-vascularity high-cellularity, low-vascularity high-cellularity, and low-vascularity low-cellularity. It was shown to be feasible to mathematically describe habitat dynamics in a preclinical model of glioma using biology-based ODEs.

Summary: The aim of this study was to evaluate the anti-glioma activity of the compound AV23 in a preclinical model of glioblastoma, as well as its effects on behavioral parameters and toxicology. The results showed that AV23 reduced tumor volume, reversed hypoglycemia and weight loss caused by glioblastoma, and restored locomotion. Serum biochemical parameters remained unchanged. These findings suggest that AV23 may be a promising candidate for the treatment of glioblastoma.

The study demonstrated that the sensitivity and specificity of MRI findings in assessing high-grade gliomas were 100% and 91% respectively, with an overall accuracy of 94.9%. MRI is considered a non-invasive method and is particularly useful when a biopsy procedure is contraindicated or rejected by the patient.
