Heterogeneity and endotypes in type 1 diabetes mellitus

Maria J. Redondo et al.

Summary: There is a recognition that T1DM is a heterogeneous disease with distinct endotypes, which has important implications for clinical practice. The current classification schema for diabetes mellitus does not capture the complexity of the disease or guide clinical management effectively. Identifying endotypes can help achieve precision medicine in diabetes mellitus and improve prediction, prevention, and treatment of T1DM.

Association between primary graft function and 5-year outcomes of islet allogeneic transplantation in type 1 diabetes: a retrospective, multicentre, observational cohort study in 1210 patients from the Collaborative Islet Transplant Registry

Mikael Chetboun et al.

Summary: This global multicentre study shows a linear and independent association between primary graft function (PGF) and 5-year clinical outcomes of islet transplantation. The study reveals that PGF is closely related to the success rate of islet transplantation, graft exhaustion, inadequate glucose control, and the need for exogenous insulin therapy. It emphasizes the importance of PGF in long-term outcomes of islet transplantation.

Glucose-6-phosphatase catalytic subunit 2 negatively regulates glucose oxidation and insulin secretion in pancreatic β-cells

Mohsin Rahim et al.

Summary: Previous studies have shown a link between the G6PC2 gene and blood glucose control. In this study, the researchers further investigated the role of G6PC2 in beta-cell metabolism. The results demonstrated that deletion of G6PC2 increased glycolytic and citric acid cycle fluxes, enhanced insulin secretion, and increased cytosolic redox potential and reductive carboxylation flux.

The PRIDE database resources in 2022: a hub for mass spectrometry-based proteomics evidences

Yasset Perez-Riverol et al.

Summary: PRIDE is the largest data repository of mass spectrometry-based proteomics data in the world, with around 500 datasets submitted per month. In addition to continuous improvements in data pipelines and infrastructure, PRIDE has developed the Spectra Archive and MAGE-TAB file format to enhance sample metadata annotation and access to mass spectra.

Islet cell dedifferentiation is a pathologic mechanism of long-standing progression of type 2 diabetes

Kikuko Amo-Shiinoki et al.

Summary: The study reveals that there is a significant presence of dedifferentiation in diabetic islets in Japanese individuals, leading to beta cell loss. As the disease progresses, the proportion of dedifferentiated cells increases substantially, indicating that islet remodeling with dedifferentiation is the underlying cause of beta cell failure during the course of diabetes progression in humans.
