Functional characterization of SGLT1 using SSM-based electrophysiology: Kinetics of sugar binding and translocation

Summary: In addition to the ongoing efforts in determining the structural information, detailed functional studies on transporters are crucial for a comprehensive understanding of the underlying transport mechanisms. Using solid supported membrane-based electrophysiology (SSME), we investigated the kinetic characteristics of the Na+/sugar cotransporter SGLT1, including sugar binding and transport, as well as the effects of different ions on these processes.

Summary: Proton-dependent oligopeptide transporters (POTs) are important for the transport of dietary peptides and peptidomimetic drugs. This study provides insights into the binding and transport of diverse peptide substrates by analyzing the X-ray structures of the bacterial POT DtpB. It is concluded that peptides with compact hydrophobic residues have the highest affinity for DtpB based on molecular docking and machine learning.

Protonation State of a Histidine Residue in Human Oligopeptide Transporter 1 (hPEPT1) Regulates hPEPT1-Mediated Efflux Activity

Summary: The study revealed that the pH-dependent efflux process mediated by hPEPT1 in CHO/hPEPT1 cells is regulated by a single amino acid residue with a pKa value around 5.7. The efflux activity by hPEPT1 decreases as extracellular pH decreases, showing that the protonation state of a histidine residue near the substrate recognition site plays a crucial role in regulating the efflux process. These findings demonstrate that hPEPT1 is a bi-directional transporter and that its efflux activity is modulated in a pH-dependent manner.

A Solid Supported Membrane-Based Technology for Electrophysical Screening of B0AT1-Modulating Compounds

Summary: Classical high-throughput screening technologies such as fluorescence-based, radioactive, and mass spectrometry (MS)-based uptake assays provide rapid results for pharmacological HTS, but can lead to high false-positive hit rates. Direct electrophysiology-based assay technologies for transporter proteins are complex and time-consuming, while solid supported membrane (SSM)-based electrophysiology offers a sensitive and membrane-based method for transporter analysis. The study demonstrated the suitability of SSM-based technique for rapid and automated identification of small molecules that can inhibit B(0)AT1 activity.
