Preparation and In Vitro Validation of a Cucurbit[7]uril-Peptide Conjugate Targeting the LDL Receptor

Xue Yang et al.

Summary: In this study, a cyclic peptide (VH4127) targeting the LDL receptor (LDLR) was noncompetitively coupled to cucurbit[7]uril (CB[7] ) to develop a new drug delivery system (DDS) named CB[7]-VH4127, which maintained its binding affinity to LDLR. The resulting supramolecular complex A680-Ada & BULL;CB[7]-VH4127 demonstrated conserved LDLR-binding potential and improved LDLR-mediated endocytosis and intracellular accumulation in LDLR-expressing cells. This new DDS opens up new possibilities for targeting and intracellular delivery to LDLR-expressing tissues or tumors.

Stimuli-responsive mechanically interlocked molecules constructed from cucurbit[n]uril homologues and derivatives

Jing-Xin Liu et al.

Summary: Since the successful separation of different cucurbit[n]uril homologues in 2001, the field of cucurbit[n]uril supramolecular chemistry has rapidly developed. The combination of Q[n] cavity size and various external stimuli has led to the creation of various Q[n]-based mechanically interlocked molecules (MIMs), including (pseudo)rotaxanes, catenanes, dendrimers, and poly(pseudo)rotaxanes. This review article highlights the important advancements in the field of Q[n]-based MIMs over the past two decades, discusses examples of heterowheel (pseudo)rotaxanes and poly(pseudo)rotaxanes involving Q[n]s, and reflects on the opportunities and challenges of constructing Q[n]-based stimuli-responsive MIMs.

Polysaccharide-based supramolecular drug delivery systems mediated via host-guest interactions of cucurbiturils

Yiyun Gao et al.

Summary: Natural polysaccharides and their derivatives have great potential in drug delivery, with reversible crosslinking by Cucurbit[n]uril showing stimulus responsiveness for constructing stimuli-responsive drug release systems. The host-guest interactions of CB[6] and CB[8] in polysaccharide-based supramolecular drug delivery systems allow for easy tuning of rheological properties and drug release profiles.

Guest Exchange by a Partial Energy Ratchet in Water

Xue Yang et al.

Summary: Chemists have successfully mimicked the function of natural molecular machines with synthetic systems in organic solvents and exchanged guest molecules in synthetic macrocycles for the production of pH-responsive hydrogels in water. Cucurbiturils may be excellent ring molecules for the construction of advanced molecular machines working in water.

Applications of Macrocyclic Host Molecules in Immune Modulation and Therapeutic Delivery

Shreya S. Soni et al.

Summary: The immune system is crucial in human disease development and progression, making modulation of the immune response a key therapeutic target. Methods of manipulating the immune system through therapeutic delivery, such as local delivery of biomaterials or systemic delivery of soluble material systems, enable control of biomaterial composition at the molecular-scale, leading to next-generation immunotherapeutics and immunotheranostics.

The Effect of Cucurbit[7]uril on the Antitumor and Immunomodulating Properties of Oxaliplatin and Carboplatin

Ekaterina Pashkina et al.

Summary: Cucurbit[7]uril (CB[7]) as a molecular container can interact with platinum(II) anticancer drugs, affecting their biological properties and antitumor effects. Encapsulation of oxaliplatin in CB[7] was found to reduce the toxicity of the drug while increasing its cytotoxic effect on tumor cell lines. In vivo, CB[7] enhanced the antitumor effect of carboplatin but also increased its acute toxicity.
